Principle of the assay: ELAM-1 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-ELAM-1 antibody and an ELAM-1-HRP conjugate. The assay sample and buffer are incubated together with ELAM-1-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the ELAM-1 concentration since ELAM-1 from samples and ELAM-1-HRP conjugate compete for the anti-ELAM-1 antibody binding site. Since the number of sites is limited, as more sites are occupied by ELAM-1 from the sample, fewer sites are left to bind ELAM-1-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The ELAM-1 concentration in each sample is interpolated from this standard curve.

NCBI Description

The protein encoded by this gene is found in cytokine-stimulated endothelial cells and is thought to be responsible for the accumulation of blood leukocytes at sites of inflammation by mediating the adhesion of cells to the vascular lining. It exhibits structural features such as the presence of lectin- and EGF-like domains followed by short consensus repeat (SCR) domains that contain 6 conserved cysteine residues. These proteins are part of the selectin family of cell adhesion molecules. Adhesion molecules participate in the interaction between leukocytes and the endothelium and appear to be involved in the pathogenesis of atherosclerosis. [provided by RefSeq, Jul 2008]
