Intended Uses: This ECF ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Porcine ECF. This ELISA kit for research use only!

Principle of the Assay: ECF ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-ECF antibody and an ECF-HRP conjugate. The assay sample and buffer are incubated together with ECF-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the ECF concentration since ECF from samples and ECF-HRP conjugate compete for the anti-ECF antibody binding site. Since the number of sites is limited, as more sites are occupied by ECF from the sample, fewer sites are left to bind ECF-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The ECF concentration in each sample is interpolated from this standard curve.

NCBI Description

This antimicrobial gene is one of several chemokine genes clustered on the q-arm of chromosome 17. Chemokines form a superfamily of secreted proteins involved in immunoregulatory and inflammatory processes. The superfamily is divided into four subfamilies based on the arrangement of the N-terminal cysteine residues of the mature peptide. This chemokine, a member of the CC subfamily, displays chemotactic activity for eosinophils, but not mononuclear cells or neutrophils. This eosinophil-specific chemokine is thought to be involved in eosinophilic inflammatory diseases such as atopic dermatitis, allergic rhinitis, asthma and parasitic infections.
