By searching databases for aminoacyl-tRNA synthetases containing a mitochondrial targeting sequence, Bonnefond et al. (2005) identified EARS2, which the called mitochondrial GLURS. The deduced 523-amino acid protein has an N-terminal mitochondrial targeting signal with a predicted cleavage site after residue 25. EARS2 has characteristics of a class I aminoacyl-tRNA synthetase, including a classical Rossmann fold.
Principle of the Assay: This assay employs a two-site sandwich ELISA to quantitate EARS2 in samples. An antibody specific for EARS2 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyEARS2 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for EARS2 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of EARS2 bound in the initial step. The color development is stopped and the intensity of the color is measured.
NCBI and Uniprot Product Information
NCBI Description
This gene encodes a member of the class I family of aminoacyl-tRNA synthetases. These enzymes play a critical role in protein biosynthesis by charging tRNAs with their cognate amino acids. This protein is encoded by the nuclear genome but is likely to be imported to the mitochondrion where it is thought to catalyze the ligation of glutamate to tRNA molecules. Mutations in this gene have been associated with combined oxidative phosphorylation deficiency 12 (COXPD12). Alternative splicing results in multiple transcript variants. Uniprot Description
EARS2: Catalyzes the attachment of glutamate to tRNA(Glu) in a two-step reaction: glutamate is first activated by ATP to form Glu-AMP and then transferred to the acceptor end of tRNA(Glu). Belongs to the class-I aminoacyl-tRNA synthetase family.

The Human EARS2 ears2 (Catalog #AAA282957) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA282957 ELISA Kit recognizes Human EARS2. It is sometimes possible for the material contained within the vial of "Probable glutamyl-tRNA synthetase, ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.Precautions
