Form/Format
The protein was lyophilized without any additives.
Sterile Filtered White lyophilized (freeze-dried) powder.
Sequence
The sequence of the first five N-terminal amino acids was determined and was found to be Met-Phe-Pro-Thr-Ser
Sequence Length
211
Solubility
It is recommended to reconstitute the lyophilized Interleukin-6 in 100mM acetic acid to 1.0mg/ml and incubated for 30 minutes at room temperature to regain full activity, which can then be further diluted to other aqueous soltutions.
Protein Content
Protein quantitation was carried out by two independent methods:1. UV spectroscopy at 280 nm using the absorbency value of 0.55 as the extinction coefficient for a 0.1% (1mg/ml) solution. This value is calculated by the PC GENE computer analysis program of protein sequences (IntelliGenetics). 2. Analysis by RP-HPLC, using a standard solution of IL-6 as a Reference Standard.
Biological Activity
The ED50 = 0.03-0.1 ng/ml corresponding to a specific activity of 10,000,000-34,000,000IU/mg. The biological activity is determined by measuring the dose-dependent proliferation of IL-6 dependent B9 cells. A concentration range of 0.1-10.0 ng/ml is effective for most in vitro applications.
Preparation and Storage
Lyophilized Interleukin-6 although stable at room temperature for 3 weeks, should be stored desiccated below -18 degree C. Upon reconstitution IL6 should be stored at 4 degree C between 2-7 days and for future use below -18 degree C.For long term storage it is recommended to add a carrier protein (0.1% HSA or BSA).Please prevent freeze-thaw cycles.
Related Product Information for rIL 6 active protein
Description: Interleukin-6 Rat Recombinant produced in E Coli is a single, non-glycosylated polypeptide chain containing 187 amino acids and having a molecular mass of 21732 Dalton. The IL6 is purified by proprietary chromatographic techniques.

Introduction: Interleukin-6 is a potent pro-inflammatory cytokine primarily produced by activated T cells and an assortment of other cells including endothelial cells and macrophages. IL-6 affects B and T lymphocytes and has been shown to have a role in host defense, acute phase reactions, immune responses and hematopoiesis.

NCBI Description

a cytokine involved in development and possibly in neurodegenerative processes [RGD, Feb 2006]

Uniprot Description

Cytokine with a wide variety of biological functions. It is a potent inducer of the acute phase response. Plays an essential role in the final differentiation of B-cells into Ig-secreting cells Involved in lymphocyte and monocyte differentiation. Acts on B-cells, T-cells, hepatocytes, hematopoietic progenitor cells and cells of the CNS. Required for the generation of T(H)17 cells. Also acts as a myokine. It is discharged into the bloodstream after muscle contraction and acts to increase the breakdown of fats and to improve insulin resistance. It induces myeloma and plasmacytoma growth and induces nerve cells differentiation ().

Product Notes

The rIL 6 il6 (Catalog #AAA142265) is an Active Protein produced from E Coli and is intended for research purposes only. The product is available for immediate purchase. The amino acid sequence is listed below: The sequence of the first five N-terminal amino acids was determined and was found to be Met-Phe-Pr o-Thr-Ser. It is sometimes possible for the material contained within the vial of "Interleukin-6, Active Protein" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
