E Coli
Purity/Purification
Greater than 98.0% as determined by:
(a) Analysis by reducing and non-reducing SDS-PAGE Silver Stained gel.
(b) Gel filtration chromatography under non-denaturing conditions.
Form/Format
Lyophylized white powder
Sequence
The sequence of the first six N-terminal amino acids was determined and was found to be Ala-Ser-Ala-Pro-Tyr-Thr.
Sequence Length
271
Solubility
It is recommended to reconstitute the lyophilized IL-1a in sterile 18Momega-cm H2O not less than 100ug/ml, which can then be further diluted to other aqueous solutions containing carrier protein such as BSA, HSA or similar.
Protein Content
Protein quantitation was carried out by UV spectroscopy at 280 nm using the absorbency value of 1.33 as the extinction coefficient for a 0.1% (1mg/ml) solution. This value is calculated by the DNAman computer analysis program of protein sequences.
Dimers and Aggregates
Less than 2% as determined by gel filtration under non-denaturing conditions.
Endotoxin
Less than 0.05 ng/ug (0.5 EU/ug) of recombinant IL-1a.
Formulation
Interleukin-1a is lyophilized from a concentrated (1mg/ml) solution in water containing NaHCO3.
Biological Activity
murine IL-1a is biologically active in helper T cell line D10.G4.1
Preparation and Storage
Lyophilized IL-1-alpha although stable at room temperature for 3 weeks, should be stored desiccated below -180 degree C. Upon reconstitution Interleukin-3 should be stored at 40C between 2-7 days and for future use below -180 degree C. For long term storage it is recommended to add a carrier protein (0.1% HSA or BSA).
Please avoid freeze-thaw cycles.
Related Product Information for mIL-1a active protein
IL-1a is a proinflammatory cytokine produced in a variety of cells including monocytes, tissue macrophages, keratinocytes and other epithelial cells. Both IL-1a and IL-1b binds to the same receptor and has similar if not identical biological properties. These cytokines have a broad range of activities including, stimulation of thymocyte proliferation, by inducing IL-2 release, B-cell maturation and proliferation, mitogenic FGF-like activity and the ability to stimulate the release of prostaglandin and collagenase from synovial cells. Recombinant mouse IL-1a is an 18.4 kDa protein containing 157 amino acid residues. Recombinant monomeric murine IL-1a is purified by proprietary chromatographic techniques.

NCBI Description

The protein encoded by this gene is a member of the interleukin 1 cytokine family. This cytokine is a pleiotropic cytokine involved in various immune responses, inflammatory processes, and hematopoiesis. This cytokine is produced by monocytes and macrophages as a proprotein, which is proteolytically processed and released in response to cell injury, and thus induces apoptosis. This gene and eight other interleukin 1 family genes form a cytokine gene cluster on chromosome 2. It has been suggested that the polymorphism of these genes is associated with rheumatoid arthritis and Alzheimer's disease. [provided by RefSeq, Jul 2008]

Uniprot Description

IL1A: Produced by activated macrophages, IL-1 stimulates thymocyte proliferation by inducing IL-2 release, B-cell maturation and proliferation, and fibroblast growth factor activity. IL-1 proteins are involved in the inflammatory response, being identified as endogenous pyrogens, and are reported to stimulate the release of prostaglandin and collagenase from synovial cells. Belongs to the IL-1 family.

Product Notes

The mIL-1a il1a (Catalog #AAA400091) is an Active Protein produced from E Coli and is intended for research purposes only. The product is available for immediate purchase. The amino acid sequence is listed below: The sequence of the first six N-terminal amino acids was determined and was found to be Ala-Ser-Al a-Pro-Tyr- Thr. It is sometimes possible for the material contained within the vial of "interleukin 1 alpha, Active Protein" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
