Reconstitution
The human GDF-11 should be reconstituted in water to a concentration of 0.1-1.0 mg/ml. Note: Keep pH below 5.0. Do not vortex. This solution can be stored at 2-8 degree C for up to 1 week. For extended storage, it is recommended it further dilute in a buffer and store in working aliquots at -20 degree C to -80 degree C.
Biological Activity
The ED50 was determined by its ability to inhibit induced alkaline phosphatase production by ATDC-5 chondrogenic cells. The expected ED50 for this effect is 0.08-0.10 ug/ml.
Preparation and Storage
The lyophilized protein is stable at room temperature for up to 1 month at 4 degree for 6 months. Working aliquots stored with a carrier protein are stable for at least 6 months at -20 degree C to -80 degree C. Avoid repeated freeze-thaw cycles.
Related Product Information for GDF-11 active protein
GDF-11 is a myostatin-homologous protein that acts as an inhibitor of nerve tissue growth. GDF-11 has been shown to suppress neurogenesis through a myostatin-like pathway, which involves arrest of progenitor cell-cycle in the G1 phase. Similarities between myostatin and GDF-11, which are 90% identical in their amino acid sequence, suggests that the regulatory mechanisms responsible for maintaining proper tissue size during neural and muscular development might be the same. Recombinant human GDF-11 is a 25.0 kDa disulfide-linked homodimer containing two 109 amino acid polypeptide chains. It is highly homologous to myostatin/GDF-8 sharing 90% amino-acid sequence identity.

NCBI Description

The protein encoded by this gene is a member of the bone morphogenetic protein (BMP) family and the TGF-beta superfamily. This group of proteins is characterized by a polybasic proteolytic processing site which is cleaved to produce a mature protein containing seven conserved cysteine residues. The members of this family are regulators of cell growth and differentiation in both embryonic and adult tissues. Studies in mice and Xenopus suggest that this protein is involved in mesodermal formation and neurogenesis during embryonic development. [provided by RefSeq, Jul 2008]

Uniprot Description

Function: Secreted signal that acts globally to specify positional identity along the anterior/posterior axis during development. Play critical roles in patterning both mesodermal and neural tissues and in establishing the skeletal pattern.

Subunit structure: Homodimer; disulfide-linked

By similarity.

Subcellular location: Secreted

Probable.

Sequence similarities: Belongs to the TGF-beta family.

Product Notes

The GDF-11 gdf11 (Catalog #AAA691735) is an Active Protein produced from E Coli and is intended for research purposes only. The product is available for immediate purchase. The Human GDF-11 reacts with Human and may cross-react with other species as described in the data sheet. The amino acid sequence is listed below: NLGLDCDEHS SESRCCRYPL TVDFEAFGWD WIIAPKRYKA NYCSGQ CEYM FMQKYPHTHL VQQANPRGSA GPCCTPTKMS PINMLYFNDK Q QIIYGKIPG MVVDRCGCS. It is sometimes possible for the material contained within the vial of "GDF-11, Active Protein" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
