Glycolate oxidase is a member of the superfamily of the alpha-hydroxy acid oxidases (HAO), enzymes that are present in both plants and animals. It catalyzes the FMN-mediated oxidation of glycolate to glyoxylate and glyoxylate to oxalate with reduction of oxygen to hydrogen peroxide. The co-factor, FMN, is tightly bound but not covalently linked to the protein. In humans and other vertebrates, HAOs are found primarily in the peroxisomes of liver, kidney, and pancreas. Three HAOs have been identified in humans. HAO-1 is most highly expressed in liver and pancreas and is most active on glycolate, two-carbon substrates. Glycolate oxidase oxidizes glycolic acid to glyoxylate, and can also oxidize glyoxylate into oxalate. These reactions are central to the toxicity of ethylene glycol poisoning. Recently, HAO has been identified as a major contributor to hyperoxaluria, a disorder in which large deposits of calcium oxalate form kidney stones.

NCBI Description

This gene is one of three related genes that have 2-hydroxyacid oxidase activity yet differ in encoded protein amino acid sequence, tissue expression and substrate preference. Subcellular location of the encoded protein is the peroxisome. Specifically, this gene is expressed primarily in liver and pancreas and the encoded protein is most active on glycolate, a two-carbon substrate. The protein is also active on 2-hydroxy fatty acids. The transcript detected at high levels in pancreas may represent an alternatively spliced form or the use of a multiple near-consensus upstream polyadenylation site. [provided by RefSeq, Jul 2008]

The immunogen sequence is aa1-370. It is sometimes possible for the material contained within the vial of "Hydroxyacid Oxidase-1/HAO-1, Active Protein" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening.
