>=90% (SDS-PAGE and RP-HPLC)
Synonyms
PPARg; Recombinant; Human (Peroxisome Proliferator Activated Receptor; gamma; HUMPPARG; NR1C3; PPARG1; PPARG2); PPARG active protein
Ordering
For Research Use Only!
Host
E Coli
Purity/Purification
Highly Purified
>=90% (SDS-PAGE and RP-HPLC)
Form/Format
Supplied as a liquid in PBS, 50% glycerol, 0.01% sodium azide.
Biological Activity
Specific activity is 3-4 units/ml obtained by measuring the increase of NADPH in absorbance at 340nm resulting from the reduction of NADP. In the coupled mode, one unit will produce 1.0 umole of NADPH per minute as glucose is phosphorylated by ATP at pH 7.4 at 30ºC
Preparation and Storage
May be stored at 4 degree C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20 degree C. Aliquots are stable for 6 months at -20 degree C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
Related Product Information for PPARG active protein
The peroxisome proliferator activated receptors (PPARs) are ligand-activated transcription factors within the nuclear receptor superfamily which play important roles in adipogenesis, glucose homeostasis and inflammation. Three different isotypes can be distinguished: alpha, beta and gamma. PPAR-g is mainly expressed in adipose tissue and to lesser extent in colon, the immune system and the retina. Whereas PPARa operates in the catabolism of fatty acids in the liver, PPARg influences the storage of fatty acids in the adipose tissue and plays a role in adipocyte differentiation. PPARg binds to DNA as a heterodimer with retinoid X receptor (RXR), activating expression of target genes after binding ligand.

Recombinant Human Peroxisome Proliferator Activated Receptor Gamma expressed in E. coli is fused to an amino terminal hexahistidine tag and has a molecular weight of 59.2kD.

Uniprot Description

Function: Nuclear receptor that binds peroxisome proliferators such as hypolipidemic drugs and fatty acids. Once activated by a ligand, the nuclear receptor binds to DNA specific PPAR response elements (PPRE) and modulates the transcription of its target genes, such as acyl-CoA oxidase. It therefore controls the peroxisomal beta-oxidation pathway of fatty acids. Key regulator of adipocyte differentiation and glucose homeostasis. ARF6 acts as a key regulator of the tissue-specific adipocyte P2 (aP2) enhancer. Acts as a critical regulator of gut homeostasis by suppressing NF-kappa-B-mediated proinflammatory responses

By similarity.

Enzyme regulation: PDPK1 activates its transcriptional activity independently of its kinase activity

By similarity.

Subunit structure: Heterodimer with other nuclear receptors, such as RXRA. The heterodimer with the retinoic acid receptor RXRA is called adipocyte-specific transcription factor ARF6. Interacts with NCOA6 coactivator, leading to a strong increase in transcription of target genes. Interacts with coactivator PPARBP, leading to a mild increase in transcription of target genes. Interacts with NOCA7 in a ligand-inducible manner. Interacts with NCOA1 and NCOA2 LXXLL motifs. Interacts with ASXL1, ASXL2, DNTTIP2, FAM120B, MAP2K1/MEK1, NR0B2, PDPK1, PRDM16, PRMT2 and TGFB1I1. Interacts (via DNA-binding domain) with HELZ2; the interaction stimulates the transcriptional activity of PPARG. Interacts (when activated by agonist) with PPP5C

By similarity.

Subcellular location: Nucleus

By similarity. Cytoplasm

By similarity. Note: Redistributed from the nucleus to the cytosol through a MAP2K1/MEK1-dependent manner

By similarity.

Tissue specificity: Highest expression in adipose tissue. Lower in spleen and lung. Also detected in ovary, mammary gland and small intestine.

Post-translational modification: Phosphorylated at basal conditions and dephosphorylated when treated with the ligand. May be dephosphorylated by PPP5C. The phosphorylated form may be inactive and dephosphorylation induces adipogenic activity

By similarity.

Sequence similarities: Belongs to the nuclear hormone receptor family. NR1 subfamily.Contains 1 nuclear receptor DNA-binding domain.

Product Notes

The PPARG pparg (Catalog #AAA635583) is an Active Protein produced from E Coli and is intended for research purposes only. The product is available for immediate purchase. It is sometimes possible for the material contained within the vial of "PPARg, Active Protein" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
