Biological Activity
Specific activity > 20 A650/cm/min/mg. Enzymatic activity was confirmed by measuring the aggregation of insulin in the presence of DTT.
Assay
1. Prepare a 180ul assay buffer into a suitable container: The concentrations are 0.67mM DTT, insulin 5.3mg, 1.2mM EDTA
2. Dilute insulin 8mg/ml in 3mM EDTA buffer (pH7.0) and incubate diluted insulin for 5minutes at 37C.
3. Dilute PDIA6 to 300ug/ml in 0.1%BSA.
4. Mix 40ul diluted PDIA6 and 10mM DTT 10ul.
5. Mix 50ul of 300ug/ml PDIA6 in 10mM DTT and 100ul of 8mg/ml insulin.
6. Incubate at 37C for 4minutes.
7. Read at 650nm in kinetic mode for 10minutes.
Preparation and Storage
Can be stored at 4 degree C short term (1-2 weeks).
For long term storage, aliquot and store at -20 degree C or -70 degree C.
Avoid repeated freezing and thawing cycles.

PDIA6 is a member of the protein disulfide isomerase (PDI). PDI is an enzyme in the endoplasmic reticulum in eukaryotes or periplasmic space of prokaryotes that catalyzes the formation and breakage of disulfide bonds between cysteine residues within proteins as they fold. PDIA6 function as a chaperone that inhibits aggregation of misfolded proteins. It plays a role in platelet aggregation and activation by agonists such as convulxin, collagen and thrombin. Recombinant human PDIA6 protein, fused to His-tag at N-terminus, was expressed in E Coli and purified by using conventional chromatography techniques.

NCBI Description

This gene encodes a member of the disulfide isomerase (PDI) family of endoplasmic reticulum (ER) proteins that catalyze protein folding and thiol-disulfide interchange reactions. The encoded protein has an N-terminal ER-signal sequence, two catalytically active thioredoxin (TRX) domains, a TRX-like domain, and a C-terminal ER-retention sequence. This protein inhibits the aggregation of misfolded proteins and exhibits both isomerase and chaperone activity. Alternative splicing results in multiple transcript variants encoding different isoforms. [provided by RefSeq, Dec 2016]

Negatively regulates the unfolded protein response (UPR) through binding to UPR sensors such as ERN1, which in turn inactivates ERN1 signaling (PubMed:24508390). May also regulate the UPR via the EIF2AK3 UPR sensor (PubMed:24508390). Plays a role in platelet aggregation and activation by agonists such as convulxin, collagen and thrombin (PubMed:15466936).
