Macrophage Inflammatory Protein 1 alpha, Rat Active Protein | CCL3 active protein

Macrophage Inflammatory Protein 1 alpha, Recombinant, Rat (MIP-1a, CCL3)

Gene Names
CCL3; MIP1A; SCYA3; G0S19-1; LD78ALPHA; MIP-1-alpha
Purity
Highly Purified
95% as determined by RP-HPLC, or SDS-PAGE.
Synonyms
Macrophage Inflammatory Protein 1 alpha; Rat; Recombinant; Rat (MIP-1a; CCL3); CCL3 active protein
Ordering
For Research Use Only!
Host
E Coli
Purity/Purification
Highly Purified
95% as determined by RP-HPLC, or SDS-PAGE.
Form/Format
Supplied as a lyophilized powder without additives.
Sequence
The sequence of the first five N-terminal amino acids: Ala-Pro-Tyr-Gly-Ala.
Biological Activity
Recombinant Rat MIP-1a is fully biologically active when compared to standard. The Activity is calculated by the ability to chemoattract of rat peritoneal macrophages using a conc. of 60 ng/ml.
Protein Content
Protein quantitation was carried out by two independent methods:
1. UV spectroscopy at 280 nm using the absorbency value of 1.38 as the extinction coefficient for a 0.1% (1mg/ml) solution. This value is calculated by the PC GENE computer analysis program of protein sequences (IntelliGenetics).
2. Analysis by RP-HPLC, using a calibrated solution of MIP-1 alpha as a Reference Standard.
Preparation and Storage
-20 degree C
Stability: Lyophilized MIP-1alpha although stable at room temperature for 3 weeks, should be stored desiccated below -180C. Upon reconstitution MIP-1 alpha should be stored at 40C between 2-7 days and for future use below -180C. For long term storage it is recommended to add a carrier protein (0.1% HSA or BSA). Please avoid freeze-thaw cycles.

NCBI Description

This locus represents a small inducible cytokine. The encoded protein, also known as macrophage inflammatory protein 1 alpha, plays a role in inflammatory responses through binding to the receptors CCR1, CCR4 and CCR5. Polymorphisms at this locus may be associated with both resistance and susceptibility to infection by human immunodeficiency virus type 1.

Uniprot Description

Function: Monokine with inflammatory and chemokinetic properties. Binds to CCR1, CCR4 and CCR5. One of the major HIV-suppressive factors produced by CD8+ T-cells. Recombinant MIP-1-alpha induces a dose-dependent inhibition of different strains of HIV-1, HIV-2, and simian immunodeficiency virus (SIV). Ref.8

Subunit structure: Self-associates. Also heterodimer of MIP-1-alpha(4-69) and MIP-1-beta(3-69). Ref.11

Subcellular location: Secreted.

Induction: By TPA or PHA (TPA = 12-O-tetradecanoyl phorbol-13 acetate (tumor promoter); PHA = phytohemagglutinin (T-cell mitogen)).

Post-translational modification: N-terminal processed form LD78-alpha(4-69) is produced by proteolytic cleavage after secretion from HTLV1-transformed T-cells.

Sequence similarities: Belongs to the intercrine beta (chemokine CC) family.

Product Notes

The CCL3 ccl3 (Catalog #AAA651047) is an Active Protein produced from E Coli and is intended for research purposes only. The product is available for immediate purchase. The amino acid sequence is listed below: The sequence of the first five N-terminal amino acids: Ala-Pro-Ty r-Gly-Ala. It is sometimes possible for the material contained within the vial of "Macrophage Inflammatory Protein 1 alpha, Rat, Active Protein" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
