Assay #1: The ED50 was determined by the dose-dependent induction of IL-6 in primary human foreskin fibroblast and is ~2ng/ml.
Assay #2: Measured by its ability to induce IL-6 production by NHDF cells.
Preparation and Storage
Lyophilized powder may be stored at -20 degree C. Stable for 12 months at -20 degree C. Reconstitute with sterile ddH2O, carrier protein. Aliquot to avoid repeated freezing and thawing. Store at -20 degree C. Reconstituted product is stable for 6 months at -20 degree C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
Related Product Information for Interleukin 17 active protein
The originally described IL-17 protein, now known as IL-17A, is a homodimer of two 136aa chains, secreted by activated T-cells that act on stromal cells to induce production of proinflammatory and hematopoietic bioactive molecules. Today, IL-17 represents a family of structurally-related cytokines that share a highly conserved C-terminal region but differ from one another in their N-terminal regions and in their distinct biological roles. The six known members of this family, IL-17A through IL-17F, are secreted as homodimers. IL-17A exhibits cross-species bioactivity between human and murine cells. Recombinant human IL-17A is a 31kD disulfide-linked homodimer of two 136aa polypeptide chains.

NCBI Description

The protein encoded by this gene is a proinflammatory cytokine produced by activated T cells. This cytokine regulates the activities of NF-kappaB and mitogen-activated protein kinases. This cytokine can stimulate the expression of IL6 and cyclooxygenase-2 (PTGS2/COX-2), as well as enhance the production of nitric oxide (NO). High levels of this cytokine are associated with several chronic inflammatory diseases including rheumatoid arthritis, psoriasis and multiple sclerosis. [provided by RefSeq, Jul 2008]

Uniprot Description

IL17A: Induces stromal cells to produce proinflammatory and hematopoietic cytokines. Enhances the surface expression of ICAM1/intracellular adhesion molecule 1 in fibroblasts. Belongs to the IL-17 family.

Protein type: Secreted, signal peptide; Secreted

Chromosomal Location of Human Ortholog: 6p12

Cellular Component: extracellular space; external side of plasma membrane

Molecular Function: cytokine activity

Biological Process: cell death; cell-cell signaling; apoptosis; positive regulation of osteoclast differentiation; positive regulation of interleukin-23 production; protein amino acid glycosylation; immune response; positive regulation of transcription from RNA polymerase II promoter; inflammatory response

Product Notes

The Interleukin 17 (Catalog #AAA650072) is an Active Protein produced from E Coli and is intended for research purposes only. The product is available for immediate purchase. The amino acid sequence is listed below: MIVKAGITIP RNPGCPNSED KNFPRTVMVN LNIHNRNTNT NPKRSSDYYN RSTSPWNLHR NEDPERYPSV IWEAKCRHLG CINADGNVDY HMNSVPIQQE ILVLRREPPH CPNSFRLEKI LVSVGCTCVT PIVHHVA. It is sometimes possible for the material contained within the vial of "Interleukin 17, Active Protein" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
