Biological Activity
Recombianant Rat Interferon gamma is fully biologically active when compared to standard. The specific activity as determined by the cytopathic affect inhibition assay with murine NIH 3T3 cells chalenged with EMC virus was < 0.1ng/ml, corresponding to a specific activity of 1x10e7Units/mg.

1% as determined by silver-stained SDS-PAGE gel analysis.
Endotoxin Level
0.1ng/ug (IEU/ug) of IFN-gamma.
Protein Content
Protein quantitation was carried out by two independent methods:
1. UV spectroscopy at 280nm. 2. Analysis by RP-HPLC, using a standard solution of IFN-gamma as a Reference Standard.
Reconstitution
It is recommended to reconstitute the lyophilized Rat Interferon-gamma in sterile 18M -cm H2O not less than 100ug/ml, which can then be further diluted to other aqueous solutions.
Preparation and Storage
Lyophilized powder may be stored at 4 degree C for short-term only. Reconstitute to nominal volume by adding sterile dH2O or buffer of choice and store at -20 degree C. Reconstituted product is stable for 12 months at -20 degree C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

NCBI Description

This gene encodes a member of the type II interferon family. The protein encoded is a soluble cytokine with antiviral, immunoregulatory and anti-tumor properties and is a potent activator of macrophages. Mutations in this gene are associated with aplastic anemia.

Function: Produced by lymphocytes activated by specific antigens or mitogens. IFN-gamma, in addition to having antiviral activity, has important immunoregulatory functions. It is a potent activator of macrophages, it has antiproliferative effects on transformed cells and it can potentiate the antiviral and antitumor effects of the type I interferons.

Subunit structure: Homodimer.

Subcellular location: Secreted.

Tissue specificity: Released primarily from activated T lymphocytes.

Post-translational modification: Proteolytic processing produces C-terminal heterogeneity, with proteins ending alternatively at Gly-150, Met-157 or Gly-161.

Involvement in disease: In Caucasians, genetic variation in IFNG is associated with the risk of aplastic anemia (AA) [

MIM:609135]. AA is a rare disease in which the reduction of the circulating blood cells results from damage to the stem cell pool in bone marrow. In most patients, the stem cell lesion is caused by an autoimmune attack. T-lymphocytes, activated by an endogenous or exogenous, and most often unknown antigenic stimulus, secrete cytokines, including IFN-gamma, which would in turn be able to suppress hematopoiesis.

Pharmaceutical use: Available under the name Actimmune (Genentech). Used for reducing the frequency and severity of serious infections associated with chronic granulomatous disease (CGD).

Sequence similarities: Belongs to the type II (or gamma) interferon family.
