Purified by ion affinity and reverse phase HPLC to 99% as determined by SDS-PAGE and HPLC.
Synonyms
HIV-1 tat; Strain IIIB; Recombinant (Human Immunodeficiency Virus-1); Strain IIIB active protein
Ordering
For Research Use Only!
Host
E Coli
Specificity
This protein binds to murine monoclonal antibodies of defined epitope specificity and human serum polyclonal antibodies in ELISA and Western ELISA.
Purity/Purification
Affinity Purified
Purified by ion affinity and reverse phase HPLC to 99% as determined by SDS-PAGE and HPLC.
Form/Format
Supplied as a liquid in 0.2M KCl, 5mM glutathione. No stabilizers or preservatives added.
Application Notes
A dose dependent response assay should be performed to determine the optimal concentration for use in specific applications. Dilute tat stock solution in saline-citrate buffer (150mM NaCl, 50mM sodium citrate, pH 6.5) immediately before use. Tat readily oxidizes in buffer solutions which may change its LTR dependent transcriptional activation activity. Transcriptional activation assays with tat are performed in 1-5ug/ml range. ELISA and Western ELISA require tat in 10-100ng protein range.
Biological Activity
The biological specificity of this protein was determined by LTR-CAT activation in scrapie-loaded HeLe Cells, and in U373 MG cells in the presence of 100uM chlorochine diphosphate. Tat concentrations in 1ug/ml range produced at least 25 fold increase in CAT activity. This protein inhibits DP4 (CD26) activity in vitro and DP4-dependent human T-cell activation in vivo.
Endotoxin Level
Less than 0.01EU/mg of protein as determined by BioWhittaker Kinetic QCL Kit.
Preparation and Storage
May be stored at 4 degree C for short-term only. For long-term storage, aliquot to avoid repeated freezing and thawing and freeze at -70 degree C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Aliquots are stable for at least 12 months.

Product Notes

The HIV-1 tat, Strain IIIB (Catalog #AAA637441) is an Active Protein produced from E Coli and is intended for research purposes only. The product is available for immediate purchase. Researchers should empirically determine the suitability of the HIV-1 tat, Strain IIIB for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "HIV-1 tat, Strain IIIB, Active Protein" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
