Biological Activity
The ED50 as determined by the dose-dependent stimulation of the proliferation of human AML5 cells is 1ng/ml corresponding to a specific activity of 1x10e6 units/mg. Flt3-ligand is fully biologically active when compared to standards.

Reconstitution
Reconstitute in sterile dH2O to a concentration of 0.1-1mg/ml. Further dilutions should be made into buffer containing carrier protein or medium containing serum.
Preparation and Storage
Lyophilized powder may be stored at 4 degree C for short-term only. For long-term storage, may be stored at -20 degree C. Reconstituted product can be stored at 4 degree C for 1 week. For long-term storage, store at -20 degree C. Aliquots are stable for at least 6 months at -20 degree C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.

Flt3 Ligand (flk2 ligand), is a growth factor that regulates proliferation of early hematopoietic cells. Flt3 Ligand binds to cells expressing the tryosine kinase receptor Flt3. Flt3 Ligand, by itself does not stimulate proliferation of early hematopoietic cells, but synergizes with other CSFs and interleukins to induce growth and differentiation. Unlike SCF, Flt 3 Ligand exerts no activity on mast cells. Multiple isoforms of Flt 3 Ligand have been identified. The predominant biologically active form is anchored to the cell surface as the extracellular domain of a transmembrane glycoprotein (209aa). The membrane-bound isoform can be proteolytically cleaved to generate a biologically active soluble isoform. Recombinant murine Flt3-Ligand is a soluble 18.6kD protein consisting of 163 amino acid residues.

Dendritic cells (DCs) provide the key link between innate and adaptive immunity by recognizing pathogens and priming pathogen-specific immune responses. FLT3LG controls the development of DCs and is particularly important for plasmacytoid DCs and CD8 (see MIM 186910)-positive classical DCs and their CD103 (ITGAE; MIM 604682)-positive tissue counterparts (summary by Sathaliyawala et al., 2010 [PubMed 20933441]).[supplied by OMIM, Jan 2011]
