Phenomenon explanation
The possible reasons that the actual band size differs from the predicted are as follows:
1. Splice variants: Alternative splicing may create different sized proteins from the same gene.
2. Relative charge: The composition of amino acids may affects the charge of the protein.
3. Post-translational modification: Phosphorylation, glycosylation, methylation etc.
4. Post-translation cleavage: Many proteins are synthesized as pro-proteins, and then cleaved to give the active form.
5. Polymerization of the target protein: Dimerization, multimerization etc.
Usage
Reconstitute in 20mM Tris, 150mM NaCl (pH8.0) to a concentration of 0.1-1.0 mg/mL. Do not vortex.
Preparation and Storage
Storage:
Avoid repeated freeze/thaw cycles.
Store at 2-8 degree C for one month.
Aliquot and store at -80 degree C for 12 months.

Stability Test:
The thermal stability is described by the loss rate. The loss rate was determined by accelerated thermal degradation test, that is, incubate the protein at 37 degree C for 48h, and no obvious degradation and precipitation were observed. The loss rate is less than 5% within the expiration date under appropriate storage condition.

NCBI Description

This gene encodes a member of the dishevelled (dsh) protein family. The vertebrate dsh proteins have approximately 40% amino acid sequence similarity with Drosophila dsh. This gene encodes a 90-kD protein that undergoes posttranslational phosphorylation to form a 95-kD cytoplasmic protein, which may play a role in the signal transduction pathway mediated by multiple Wnt proteins. The mechanisms of dishevelled function in Wnt signaling are likely to be conserved among metazoans. [provided by RefSeq, Jul 2008]

Uniprot Description

DVL2: a protein in the WNT/planar cell polarity (PCP) signaling pathway. Dvl2-deficient mice indicate that Dvl2 is essential for normal cardiac morphogenesis, somite segmentation and neural tube closure. Transduces the Wnt signal by interacting with the cytoplasmic Axin complex. Dvl and Axin each contain a DIX domains, which mediate their dynamic polymerization. The Dvl-Axin interaction is essential for Wnt signaling. Interacts through its PDZ domain with the C-terminal regions of VANGL1 and VANGL2. Interacts with Dixin and Rac. There is functional redundancy between Dvl1 and Dvl2 in some phenotypes.

Cellular Component: apical part of cell; cytoplasm; plasma membrane; cytoplasmic vesicle; clathrin-coated endocytic vesicle; cytosol; nucleus

Product Notes

The DVL2 dvl2 (Catalog #AAA2018666) is a Recombinant Protein produced from E.coli and is intended for research purposes only. The product is available for immediate purchase. The Recombinant Dishevelled, Dsh Homolog 2 (DVL2) reacts with Homo sapiens (Human) and may cross-react with other species as described in the data sheet. AAA Biotech's Dishevelled, Dsh Homolog 2 (DVL2) can be used in a range of immunoassay formats including, but not limited to, Positive Control; Immunogen; SDS-PAGE; Western Blot (WB). (May be suitable for use in other assays to be determined by the end user.). Researchers should empirically determine the suitability of the DVL2 dvl2 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "Dishevelled, Dsh Homolog 2 (DVL2), Recombinant Protein" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
