The deduced 929-amino acid protein contains a C-terminal kinase domain, which shares homology with the kinase domain of RIPK family members. Northern blot analysis of human tissues detected weak expression in heart, brain, placenta, skeletal muscle, kidney, pancreas, and testis.
DSTYK failed to activate NF-kappa-B (NFKB1) in a reporter gene assay. Overexpression of DSTYK induced DNA fragmentation and cell death in a dose-dependent manner with cell death occurring approximately 30 hours after transfection. Caspase inhibitory protein and morphologic studies showed that DSTYK induced both caspase-dependent and caspase-independent cell death, and both the N- and C-terminal DSTYK deletion mutants retained the ability to induce cell death.

Principle of the Assay: This assay employs a two-site sandwich ELISA to quantitate DSTYK in samples. An antibody specific for DSTYK has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyDSTYK present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for DSTYK is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of DSTYK bound in the initial step. The color development is stopped and the intensity of the color is measured.
