This assay has high sensitivity and excellent specificity for detection of DPD. No significant cross-reactivity or interference between DPD and analogues was observed. NOTE: Limited by current skills and knowledge, it is impossible for us to complete the cross-reactivity detection between DPD and all the analogues, therefore, cross reaction may still exist in some cases.

Intended Uses: This DPD ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Mouse DPD. This ELISA kit for research use only, not for therapeutic or test applications!

Principle of the Assay: DPD ELISA kit applies the competitive enzyme immunoassay technique utilizing a polyclonal anti-DPD antibody and an DPD-HRP conjugate. The assay sample and buffer are incubated together with DPD-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the DPD concentration since DPD from samples and DPD-HRP conjugate compete for the anti-DPD antibody binding site. Since the number of sites is limited, as more sites are occupied by DPD from the sample, fewer sites are left to bind DPD-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The DPD concentration in each sample is interpolated from this standard curve.

NCBI Description

The protein encoded by this gene is a pyrimidine catabolic enzyme and the initial and rate-limiting factor in the pathway of uracil and thymidine catabolism. Mutations in this gene result in dihydropyrimidine dehydrogenase deficiency, an error in pyrimidine metabolism associated with thymine-uraciluria and an increased risk of toxicity in cancer patients receiving 5-fluorouracil chemotherapy. Two transcript variants encoding different isoforms have been found for this gene. [provided by RefSeq, May 2009]
