DPF1 is a member of the d4 domain family, characterized by a zinc finger-like structural motif. This protein functions as a transcription factor which is necessary for the apoptotic response following deprivation of survival factors. It likely serves a regulatory role in rapid hematopoietic cell growth and turnover. Chestkov et al. (1996) described human DPF1, which they called Neuro-D4, a conserved gene encoding a 353-amino acid neural-specific zinc finger protein. A cysteine- and histidine-rich region of the C terminus distinguishes members of the D4 family, including Neuro-D4, from other zinc finger proteins. The human and rat Neuro-D4 cDNAs are 93% identical, and there are only 3 amino acid differences between the proteins. Different alternatively spliced cDNAs were observed.

Principle of the Assay: This assay employs a two-site sandwich ELISA to quantitate DPF1 in samples. An antibody specific for DPF1 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyDPF1 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for DPF1 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of DPF1 bound in the initial step. The color development is stopped and the intensity of the color is measured.

Uniprot Description

DPF1: May have an important role in developing neurons by participating in regulation of cell survival, possibly as a neurospecific transcription factor. Belongs to the neuron-specific chromatin remodeling complex (nBAF complex). During neural development a switch from a stem/progenitor to a post-mitotic chromatin remodeling mechanism occurs as neurons exit the cell cycle and become committed to their adult state. The transition from proliferating neural stem/progenitor cells to post-mitotic neurons requires a switch in subunit composition of the npBAF and nBAF complexes. As neural progenitors exit mitosis and differentiate into neurons, npBAF complexes which contain ACTL6A/BAF53A and PHF10/BAF45A, are exchanged for homologous alternative ACTL6B/BAF53B and DPF1/BAF45B or DPF3/BAF45C subunits in neuron-specific complexes (nBAF). The npBAF complex is essential for the self-renewal/proliferative capacity of the multipotent neural stem cells. The nBAF complex along with CREST plays a role regulating the activity of genes essential for dendrite growth. Belongs to the requiem/DPF family. 3 isoforms of the human protein are produced by alternative splicing.
