Principle of the Assay: DCAL-2 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-DCAL-2 antibody and an DCAL-2-HRP conjugate. The assay sample and buffer are incubated together with DCAL-2-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the DCAL-2 concentration since DCAL-2 from samples and DCAL-2-HRP conjugate compete for the anti-DCAL-2 antibody binding site. Since the number of sites is limited, as more sites are occupied by DCAL-2 from the sample, fewer sites are left to bind DCAL-2-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The DCAL-2 concentration in each sample is interpolated from this standard curve.

NCBI Description

This gene encodes a member of the C-type lectin/C-type lectin-like domain (CTL/CTLD) superfamily. The encoded glycoprotein is a small type II membrane receptor with an extracellular C-type lectin-like domain fold and a cytoplasmic domain with an immunoreceptor tyrosine-based activation motif. It functions as a pattern-recognition receptor that recognizes a variety of beta-1,3-linked and beta-1,6-linked glucans from fungi and plants, and in this way plays a role in innate immune response. Alternate transcriptional splice variants, encoding different isoforms, have been characterized. This gene is closely linked to other CTL/CTLD superfamily members on chromosome 12p13 in the natural killer gene complex region. [provided by RefSeq, Jul 2008]

Mediates production of reactive oxygen species in the cell. Mediates phagocytosis of C.albicans conidia. Binds T-cells in a way that does not involve their surface glycans and plays a role in T-cell activation. Stimulates T-cell proliferation. Up-regulated during differentiation from monocytes into dendritic cells. Isoform 5 interacts with RANBP9. 8 isoforms of the human protein are produced by alternative splicing.
