Principle of the Assay: CYTB5 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-CYTB5 antibody and an CYTB5-HRP conjugate. The assay sample and buffer are incubated together with CYTB5-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the CYTB5 concentration since CYTB5 from samples and CYTB5-HRP conjugate compete for the anti-CYTB5 antibody binding site. Since the number of sites is limited, as more sites are occupied by CYTB5 from the sample, fewer sites are left to bind CYTB5-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The CYTB5 concentration in each sample is interpolated from this standard curve.

NCBI Description

The protein encoded by this gene is a membrane-bound cytochrome that reduces ferric hemoglobin (methemoglobin) to ferrous hemoglobin, which is required for stearyl-CoA-desaturase activity. Defects in this gene are a cause of type IV hereditary methemoglobinemia. Three transcript variants encoding different isoforms have been found for this gene. [provided by RefSeq, Jun 2010]

CYB5A: Cytochrome b5 is a membrane bound hemoprotein which function as an electron carrier for several membrane bound oxygenases. Defects in CYB5A are the cause of methemoglobinemia CYB5A-related (METHB-CYB5A). A form of methemoglobinemia, a hematologic disease characterized by the presence of excessive amounts of methemoglobin in blood cells, resulting in decreased oxygen carrying capacity of the blood, cyanosis and hypoxia. Belongs to the cytochrome b5 family. 2 isoforms of the human protein are produced by alternative splicing.
