Principle of the Assay: The LumiAb Human Follistatin Chemiluminescent ELISA Kit contains the components necessary for quantitative determination of natural or recombinant Human Follistatin concentrations within any experimental sample including cell lysates, serum and plasma. This particular immunoassay utilizes the quantitative technique of a "Sandwich" Enzyme-Linked Immunosorbent Assay (ELISA) where the target protein (antigen) is bound in a "sandwich" format by the primary capture antibodies coated to each well-bottom and the secondary detection antibodies added subsequently by the investigator. The capture antibodies coated to the bottom of each well are specific for a particular epitope on Human Follistatin while the user-added detection antibodies bind to epitopes on the captured target protein. Amid each step of the procedure, a series of wash steps must be performed to ensure the elimination of non-specific binding between proteins to other proteins or to the solid phase. After incubation and "sandwiching" of the target antigen, a peroxidase enzyme is conjugated to the constant heavy chain of the secondary antibody (either covalently or via Avidin/Streptavidin-Biotin interactions), allowing for a sensitive luminescent reaction to ensue upon substrate addition. When the Peroxide Enhancer solution is added, the reaction catalyzed by peroxidase yields light that is representative of the antigen concentration. After a brief incubation, the microplate can be read with a luminometer, allowing for generation of a standard curve and subsequent determination of protein concentration!!Background/Introduction: Human Follistatin or FS, also known as Activin-Binding Protein is a single-chain gonadal protein that specifically inhibits pituitary follicle-stimulating hormone (FSH) release. The single FST gene encodes two isoforms, FST317 and FST344 containing 317 and 344 amino acid residues respectively, resulting from alternative splicing of the precursor mRNA. Isoform 1 is the predominant isoform in serum but is undetectable in follicular fluid. Moreover, the FST gene is located at locus 5q11.2 on chromosome 5. The monomeric protein binds directly to Activin and functions as an Activin antagonist. In regards to the primary sequence, Follistatin contains 3 follistatin-like domains, 3 Kazal-like domains and a TB (TBF-beta binding) domain. Typical modifications to the protein include 2 potential glycosylation sites at the 124th or 288th residue along with multiple sites for disulfide bridge formation.

NCBI Description

This gene belongs to the CX3C subgroup of chemokines, characterized by the number of amino acids located between the conserved cysteine residues. This is the only member of the CX3C subgroup, which contains three amino acids between cysteine residues, resulting in a Cys-X-X-X-Cys configuration. The encoded protein contains an extended mucin-like stalk with a chemokine domain on top, and exists in both a membrane-anchored form where it acts as a binding molecule, or, in soluble form, as a chemotactic cytokine. The mature form of this protein can be cleaved at the cell surface, yielding different soluble forms that can interact with the G-protein coupled receptor, C-X3-C motif chemokine receptor 1 gene product. This gene plays a role in a wide range of diseases, including cancer, vasculitis, neuropathies, atherosclerosis, inflammatory diseases, and in human immunodeficiency virus infections. Uniprot Description

CX3CL1: The soluble form is chemotactic for T-cells and monocytes, but not for neutrophils. The membrane-bound form promotes adhesion of those leukocytes to endothelial cells. May play a role in regulating leukocyte adhesion and migration processes at the endothelium. Binds to CX3CR1. Belongs to the intercrine delta family.

Product Notes

The Human CX3CL1 cx3cl1 (Catalog #AAA9510969) is a CLIA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA9510969 CLIA Kit recognizes Human CX3CL1. It is sometimes possible for the material contained within the vial of "Fractalkine, CLIA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
