Principle of the Assay: COL17 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-COL17 antibody and an COL17-HRP conjugate. The assay sample and buffer are incubated together with COL17-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the COL17 concentration since COL17 from samples and COL17-HRP conjugate compete for the anti-COL17 antibody binding site. Since the number of sites is limited, as more sites are occupied by COL17 from the sample, fewer sites are left to bind COL17-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The COL17 concentration in each sample is interpolated from this standard curve.

NCBI Description

This gene encodes the alpha chain of type XVII collagen. Unlike most collagens, collagen XVII is a transmembrane protein. Collagen XVII is a structural component of hemidesmosomes, multiprotein complexes at the dermal-epidermal basement membrane zone that mediate adhesion of keratinocytes to the underlying membrane. Mutations in this gene are associated with both generalized atrophic benign and junctional epidermolysis bullosa. Two homotrimeric forms of type XVII collagen exist. The full length form is the transmembrane protein. A soluble form, referred to as either ectodomain or LAD-1, is generated by proteolytic processing of the full length form. Defects in COL17A1 are a cause of generalized atrophic benign epidermolysis bullosa (GABEB). GABEB is a non- lethal, adult form of junctional epidermolysis bullosa characterized by life-long blistering of the skin, associated with hair and tooth abnormalities. 2 isoforms of the human protein are produced by alternative splicing.
