Intended Uses||This CY17 ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Bovine CY17. This ELISA kit for research use only!

Principle of the Assay: CY17 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-CY17 antibody and an CY17-HRP conjugate. The assay sample and buffer are incubated together with CY17-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the CY17 concentration since CY17 from samples and CY17-HRP conjugate compete for the anti-CY17 antibody binding site. Since the number of sites is limited, as more sites are occupied by CY17 from the sample, fewer sites are left to bind CY17-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The CY17 concentration in each sample is interpolated from this standard curve.

This gene encodes the type I intermediate filament chain keratin 17, expressed in nail bed, hair follicle, sebaceous glands, and other epidermal appendages. Mutations in this gene lead to Jackson-Lawler type pachyonychia congenita and steatocystoma multiplex. [provided by RefSeq, Aug 2008]

K17: a type I cytoskeletal keratin. The keratins are intermediate filament proteins responsible for the structural integrity of epithelial cells and are subdivided into cytokeratins and hair keratins. There are two types of cytoskeletal and microfibrillar keratin: type I (acidic; 40-55 kDa) [K9 to K20] and type II (neutral to basic; 56-70 kDa) [K1 to K8]. Both a basic and an acidic keratin are required for filament assembly. Generally associates with K6.
