Kit Components
Chromogenic Reagent A (BCIP/NBT, 20×) - 3 ml
Chromogenic Reagent B (Tris solution, pH9.4, 20×) - 3 ml

Introduction
1. Alkaline Phosphatase is one of the most important makers in immunology and molecular biology. Its insoluble chromogenic substrate mainly contains naphthol phosphate and BCIP. With NBT, BCIP can forms the insoluble dark blue or even bluish violet NBT-formazan, can be used for IHC and various kinds of immunospot assays.
2. This kit can be used for alkaline phosphatase coloration of cells or tissues, including identification of induced plurpotent stem cells, membrane coloration combining with alkaline phosphatase in WB and endogenic alkaline phosphatase coloration of cells or tissues.
3. For IHC or ISH, if each section needs 0.2ml of such reagent, this kit can be enough for 500 sections.

Protocol
1. After the peroxisase detection, wash with TBS thoroughly for 4 times, 5 min for each. Phosphate buffer can not be used here!
2. Add one drop of Chromogenic Reagent A and one drop of Chromogenic Reagent B into 1ml of distilled water, mix thoroughly, then, add this mixed reagent in the sample. Colorate in dark for 10-30 min. If there is no background, keep coloration until get the satisfied results.
3. Wash wish distilled water to stop the reaction. If necessary, counterstain with nuclear fast red.
4. Seal the slide / section and observe with results by microscope.

Preparation and Storage
Store at 4°C for frequent use, or store at -20°C for long term.
