Related Product Information for BMP-2 active protein
BMPs (Bone Morphogenetic Proteins) belong to the TGF-beta superfamily of structurally related signaling proteins. BMP-2 is a potent osteoinductive cytokine, capable of inducing bone and cartilage formation in association with an osteoconductive carrier such as collagen and synthetic hydroxyapatite. In addition to its osteogenic activity, BMP-2 appears to play an important role in cardiac morphogenesis, and is expressed in a variety of other tissues, including lung, liver, spleen, prostate, ovary, and small intestine. The functional form of BMP-2 is a 26 kDa protein composed of two identical 114 amino acid polypeptide chains (monomers) linked by a single disulfide bond. Each BMP-2 monomer is expressed as the C-terminal part of a precursor polypeptide, which also contains a 23 amino acid signal sequence for secretion, and a 259 amino acid propeptide. After dimerization of this precursor, the covalent bonds between the propeptide (which is also a disulfide-linked homodimer) and the mature BMP-2 ligand are cleaved by a furin-type protease. Recombinant Human BMP-2 derived from CHO cells is a homodimeric glycoprotein that consists of two 114 amino acid polypeptide chains linked by a single disulfide bond. Due to glycosylation, CHO cell-derived Human BMP-2 migrates at an apparent molecular weight of approximately 28-29 kDa by SDS-PAGE analysis under non-reducing conditions.

NCBI Description

This gene encodes a secreted ligand of the TGF-beta (transforming growth factor-beta) superfamily of proteins. Ligands of this family bind various TGF-beta receptors leading to recruitment and activation of SMAD family transcription factors that regulate gene expression. The encoded preproprotein is proteolytically processed to generate each subunit of the disulfide-linked homodimer, which plays a role in bone and cartilage development. Duplication of a regulatory region downstream of this gene causes a form of brachydactyly characterized by a malformed index finger and second toe in human patients. [provided by RefSeq, Jul 2016]

Uniprot Description

Induces cartilage and bone formation (PubMed:3201241). Stimulates the differentiation of myoblasts into osteoblasts via the EIF2AK3-EIF2A- ATF4 pathway. BMP2 activation of EIF2AK3 stimulates phosphorylation of EIF2A which leads to increased expression of ATF4 which plays a central role in osteoblast differentiation. In addition stimulates TMEM119, which upregulates the expression of ATF4 (PubMed:24362451).

Product Notes

The BMP-2 bmp2 (Catalog #AAA692291) is an Active Protein produced from CHO cells and is intended for research purposes only. The product is available for immediate purchase. The Human BMP-2 reacts with Human and may cross-react with other species as described in the data sheet. The amino acid sequence is listed below: QAKHKQRKRL KSSCKRHPLY VDFSDVGWND WIVAPPGYHA FYCHGE CPFP LADHLNSTNH AIVQTLVNSV NSKIPKACCV PTELSAISML Y LDENEKVVL KNYQDMVVEG CGCR. It is sometimes possible for the material contained within the vial of "BMP-2, Active Protein" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
