• Source Note: Optimized DNA sequence encoding Human THBD mature chain was expressed in CHO. Recombinant Human Thrombomodulin encodes amino acid residues 19-575.
• Biological Activity: Measured by its ability to activate protein C induced cleavage of the chromogenic substrate, BOC-Asp-Pro Arg-AMC in the presence of thrombin. The specific activity is greater than 500 pmoles/min/ug.
• Endotoxin: Endotoxin content was assayed using a LAL gel clot method. Endotoxin level was found to be less than 0.1 ng/ug(1EU/ug).
• Reconstitution: A quick spin of the vial followed by reconstitution in distilled water to a concentration not less than 0.1 mg/mL. This solution can then be diluted into other buffers.
• Preparation and Storage: The lyophilized protein is stable for at least 2 years from date of receipt at -20 degree C. Upon reconstitution, this product can be stored in working aliquots at 2 degree C - 8 degree C for one month, or at -20 degree C for six months, with a carrier protein without detectable loss of activity. Avoid repeated freeze/thaw cycles.

NCBI Description

The protein encoded by this intronless gene is an endothelial-specific type I membrane receptor that binds thrombin. This binding results in the activation of protein C, which degrades clotting factors Va and VIIIa and reduces the amount of thrombin generated. Mutations in this gene are a cause of thromboembolic disease, also known as inherited thrombophilia. [provided by RefSeq, Jul 2008]

Uniprot Description

Thrombomodulin is a specific endothelial cell receptor that forms a 1:1 stoichiometric complex with thrombin. This complex is responsible for the conversion of protein C to the activated protein C (protein Ca). Once evolved, protein Ca scissions the activated cofactors of the coagulation mechanism, factor Va and factor VIIIa, and thereby reduces the amount of thrombin generated.
