For Research Use Only!
Reactivity
Chicken
Specificity
This assay has high sensitivity and excellent specificity for detection of BHMT. No significant cross-reactivity or interference between BHMT and analogues was observed. NOTE: Limited by current skills and knowledge, it is impossible for us to complete the cross-reactivity detection between BHMT and all the analogues, therefore, cross reaction may still exist in some cases.
Samples
Serum, plasma, cell culture supernatants, body fluid and tissue homogenate
Assay Type
Quantitative Competitive
Sensitivity
1.0 pg/mL
Preparation and Storage
Store all reagents at 2-8 degree C.

Intended Uses: This BHMT ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Chicken BHMT. This ELISA kit for research use only, not for therapeutic or test applications!

Principle of the Assay: BHMT ELISA kit applies the competitive enzyme immunoassay technique utilizing an anti-BHMT antibody and an BHMT-HRP conjugate. The assay sample and buffer are incubated together with BHMT-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the BHMT concentration since BHMT from samples and BHMT-HRP conjugate compete for the anti-BHMT antibody binding site. Since the number of sites is limited, as more sites are occupied by BHMT from the sample, fewer sites are left to bind BHMT-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The BHMT concentration in each sample is interpolated from this standard curve.

NCBI Description

This gene encodes a cytosolic enzyme that catalyzes the conversion of betaine and homocysteine to dimethylglycine and methionine, respectively. Defects in this gene could lead to hyperhomocyst(e)inemia, but such a defect has not yet been observed. [provided by RefSeq, Jul 2008]

Uniprot Description

BHMT: Involved in the regulation of homocysteine metabolism. Converts betaine and homocysteine to dimethylglycine and methionine, respectively. This reaction is also required for the irreversible oxidation of choline.

Product Notes

The Chicken BHMT bhmt (Catalog #AAA7224784) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA7224784 ELISA Kit recognizes Chicken BHMT. It is sometimes possible for the material contained within the vial of "Betaine--homocysteine S-methyltransferase 1 (BHMT), ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
