Scientific Background: PCTK1 or Pctaire protein kinase 1 belongs to the cdc2/cdkx subfamily of the ser/thr family of protein kinases which play a main role in signal transduction cascades in terminally differentiated cells, in exocytosis and in transport of secretory cargo from the endoplasmic reticulum. PCTK1 is ubiquitously expressed with the highest levels detected in the brain and testis (1). PCTK1 maps distal to the t(X;18) synovial sarcoma breakpoint in Xp11.23. A 420-kb YAC clone positive for PCTK1 also contained the gene coding for ubiquitin-activating enzyme UBE1 previously mapped to Xp11.3. (2). PCTK1 is thought to escape X inactivation.

NCBI and Uniprot Product Information
NCBI Description
The protein encoded by this gene belongs to the cdc2/cdkx subfamily of the ser/thr family of protein kinases. It may play a role in signal transduction cascades in terminally differentiated cells; in exocytosis; and in transport of secretory cargo from the endoplasmic reticulum. This gene is thought to escape X inactivation. Alternative splicing results in multiple transcript variants encoding different isoforms. [provided by RefSeq, Dec 2009]
Uniprot Description
Function: Protein kinase that plays a role in vesicle-mediated transport processes and exocytosis. Regulates GH1 release by brain neurons. Phosphorylates NSF, and thereby regulates NSF oligomerization. Required for normal spermatogenesis. Regulates neuron differentiation and dendrite development
By similarity. Plays a role in the regulation of insulin secretion in response to changes in blood glucose levels. Can phosphorylate CCNY at 'Ser-336' (in vitro). Ref.14 Ref.15 Ref.16
Catalytic activity: ATP + a protein = ADP + a phosphoprotein. Ref.14 Ref.15 Ref.16
Subunit structure: Found in a complex containing CABLES1, CDK17 and TDRD7. Interacts with YWHAH, YWHAQ and YWHAZ. Interacts with NSF. Identified in a complex with NSF, syntaxin-1, synaptotagmin, SYN1, SYP and CDK5R1
By similarity. Interacts with BRSK2. Interacts with CCNY; this increases the CDK16 kinase activity. Ref.14 Ref.15 Ref.16
Subcellular location: Cytoplasm. Cytoplasmic vesicle › secretory vesicle. Cell membrane; Peripheral membrane protein; Cytoplasmic side. Cell junction › synapse › synaptosome
By similarity. Note: Colocalizes with insulin in pancreas islets. Recruited to the cell membrane by CCNY. Ref.15 Ref.16
Tissue specificity: Detected in pancreas islets (at protein level). Detected in brain and pancreas. Ref.15
Post-translational modification: Phosphorylation at Ser-153 inhibits kinase activity.
Sequence similarities: Belongs to the protein kinase superfamily. CMGC Ser/Thr protein kinase family. CDC2/CDKX subfamily.Contains 1 protein kinase domain.
Sequence caution: The sequence AAH01048.1 differs from that shown. Reason: Erroneous initiation. Translation N-terminally extended. The sequence AAH15607.1 differs from that shown. Reason: Erroneous initiation. Translation N-terminally extended.

Product Notes
The PCTK1 cdk16 (Catalog #AAA515158) is a Recombinant Protein produced from Sf9 insect cells and is intended for research purposes only. The product is available for immediate purchase. The tag for this protein is GST tag!!Expression System||Sf9 insect cells using baculovirus!!Source Note||Recombinant human PCTK1(107-end) was expressed by baculovirus in Sf9 insect cells. AAA Biotech's PCTK1 can be used in a range of immunoassay formats including, but not limited to, Western Blot (WB). Researchers should empirically determine the suitability of the PCTK1 cdk16 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "PCTK1, Recombinant Protein" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.Precautions
