Intended Uses: This IMA ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Human IMA. This ELISA kit for research use only!

Principle of the Assay: IMA ELISA kit applies the competitive enzyme immunoassay technique utilizing a polyclonal anti-IMA antibody and an IMA-HRP conjugate. The assay sample and buffer are incubated together with IMA-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the IMA concentration since IMA from samples and IMA-HRP conjugate compete for the anti-IMA antibody binding site. Since the number of sites is limited, as more sites are occupied by IMA from the sample, fewer sites are left to bind IMA-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The IMA concentration in each sample is interpolated from this standard curve.

NCBI Description

This gene is one of several chemokine genes clustered on the q-arm of chromosome 17. Chemokines form a superfamily of secreted proteins involved in immunoregulatory and inflammatory processes. The superfamily is divided into four subfamilies based on the arrangement of the N-terminal cysteine residues of the mature peptide. This chemokine, a member of the CC subfamily, functions as a chemoattractant for blood monocytes, memory T helper cells and eosinophils. It causes the release of histamine from basophils and activates eosinophils. This cytokine is one of the major HIV-suppressive factors produced by CD8+ cells. It functions as one of the natural ligands for the chemokine receptor chemokine (C-C motif) receptor 5 (CCR5), and it suppresses in vitro replication of the R5 strains of HIV-1, which use CCR5 as a coreceptor. Alternative splicing results in multiple transcript variants that encode different isoforms. [provided by RefSeq, Jul 2013]

The processed form RANTES(3-68) acts as a natural chemotaxis inhibitor and is a more potent inhibitor of HIV-1-infection. The second processed form RANTES(4-68) exhibits reduced chemotactic and HIV-suppressive activity compared with RANTES(1-68) and RANTES(3-68) and is generated by an unidentified enzyme associated with monocytes and neutrophils.
