IL8
Host Note
Optimized DNA sequence encoding Human HCC-1 (CCL14) mature chain was expressed in Escherichia Coli.
Endotoxin
Endotoxin content was assayed using a LAL gel clot method.
Endotoxin level was found to be less than 0.1 ng/ug (1EU/ug).
Reconstitution
A quick spin of the vial followed by reconstitution in distilled water to a concentration not less than 0.1 mg/mL. This solution can then be diluted into other buffers.
Molecular Weight Note
Human HCC-1, generated by the proteolytic removal of the signal peptide
and propeptide and has a calculated molecular mass of approximately 9 kDa.

Recombinant HCC-1 is a disulfide-linked monomeric protein consisting of 73 amino acid residue subunits and migrates as an approximately 9 kDa protein under non-reducing conditions and reducing conditions in SDS-PAGE.
Preparation and Storage
The lyophilized protein is stable for at least 2 years from date of receipt at -20 degree C.
Upon reconstitution, this cytokine can be stored in working aliquots at 2 degree - 8 degree C for one month, or at -20 degree C for six months, with a carrier protein without detectable loss of activity.

Avoid repeated freeze/thaw cycles.
Related Product Information for HCC-1 recombinant protein
Hemofiltrate C-C chemokine (HCC) -1 is a recently described monocyte chemoattractant. CCL14 (also known as HCC-1) belongs to the CC chemokine family. Its mature propeptide is a low- affinity agonist of CCR1 that is converted to a high- affinity agonist of CCR1 and CCR5 on proteolytic processing by serine proteases. Determination of the amino acid sequence of HCC-1 revealed four cysteine residues in positions characteristic of the C-C chemokine family, and comparison with the sequences of other chemokines revealed that HCC-1 was most homologous to MIP-1a. However, several functional properties of HCC-1 were atypical of chemokines. Unlike other chemokines, HCC-1 was expressed constitutively in a number of tissues and was present at high concentrations in normal human plasma. In addition, HCC-1 was reported not to be chemotactic for leukocytes.

NCBI Description

This gene, chemokine (C-C motif) ligand 14, is one of several CC cytokine genes clustered on 17q11.2. The CC cytokines are secreted proteins characterized by two adjacent cysteines. The cytokine encoded by this gene induces changes in intracellular calcium concentration and enzyme release in monocytes. Multiple transcript variants encoding different isoforms have been found for this gene. Read-through transcripts are also expressed that include exons from the upstream cytokine gene, chemokine (C-C motif) ligand 15, and are represented as GeneID: 348249. [provided by RefSeq, Dec 2009]

Uniprot Description

CCL14: Has weak activities on human monocytes and acts via receptors that also recognize MIP-1 alpha. It induced intracellular Ca(2+) changes and enzyme release, but no chemotaxis, at concentrations of 100-1,000 nM, and was inactive on T-lymphocytes, neutrophils, and eosinophil leukocytes. Enhances the proliferation of CD34 myeloid progenitor cells. The processed form HCC-1(9-74) is a chemotactic factor that attracts monocytes eosinophils, and T-cells and is a ligand for CCR1, CCR3 and CCR5. Belongs to the intercrine beta (chemokine CC) family. 2 isoforms of the human protein are produced by alternative splicing.

Product Notes

The HCC-1 ccl14 (Catalog #AAA553001) is a Recombinant Protein produced from E Coli and is intended for research purposes only. The product is available for immediate purchase. The amino acid sequence is listed below: MKISVAAIPF FLLITIALGT KTESSSRGPY HPSECCFTYT TYKIPRQRIM DYYETNSQCS KPGIVFITKR GHSVCTNPSD KWVQDYIKDM KEN. It is sometimes possible for the material contained within the vial of "HCC-1 (CCL14), Recombinant Protein" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
