Intended Uses||This CTSD ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Rat CTSD. This ELISA kit for research use only!

Principle of the Assay: CTSD ELISA kit applies the quantitative sandwich enzyme immunoassay technique. The microtiter plate has been pre-coated with a monoclonal antibody specific for CTSD. Standards or samples are then added to the microtiter plate wells and CTSD if present, will bind to the antibody pre-coated wells. In order to quantitatively determine the amount of CTSD present in the sample, a standardized preparation of horseradish peroxidase (HRP)-conjugated polyclonal antibody, specific for CTSD are added to each well to "sandwich” the CTSD immobilized on the plate. The microtiter plate undergoes incubation, and then the wells are thoroughly washed to remove all unbound components. Next, substrate solutions are added to each well. The enzyme (HRP) and substrate are allowed to react over a short incubation period. Only those wells that contain CTSD and enzyme-conjugated antibody will exhibit a change in color. The enzyme-substrate reaction is terminated by addition of a sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450 nm. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The CTSD concentration in each sample is interpolated from this standard curve.

NCBI Description

This gene encodes a lysosomal aspartyl protease composed of a dimer of disulfide-linked heavy and light chains, both produced from a single protein precursor. This proteinase, which is a member of the peptidase C1 family, has a specificity similar to but narrower than that of pepsin A. Transcription of this gene is initiated from several sites, including one which is a start site for an estrogen-regulated transcript. Mutations in this gene are involved in the pathogenesis of several diseases, including breast cancer and possibly Alzheimer disease. [provided by RefSeq, Jul 2008]

Catalytic activity: Specificity similar to, but narrower than, that of pepsin A. Does not cleave the 4-Gln-|-His-5 bond in B chain of insulin.

Subunit structure: Consists of a light chain and a heavy chain.

Subcellular location: Lysosome. Melanosome. Secreted › extracellular space. Note: Identified by mass spectrometry in melanosome fractions from stage I to stage IV. In aortic samples, detected as an extracellular protein loosely bound to the matrix (Ref.17). Ref.10 Ref.14 Ref.17

Tissue specificity: Expressed in the aorta extrcellular space (at protein level). Ref.17

Post-translational modification: N- and O-glycosylated. Ref.11 Ref.19

Polymorphism: The Val-58 allele is significantly overrepresented in demented patients (11.8%) compared with non-demented controls (4.9%). Carriers of the Val-58 allele have a 3.1-fold increased risk for developing AD than non-carriers.

Involvement in disease: Neuronal ceroid lipofuscinosis 10 (CLN10) [MIM:610127]: A form of neuronal ceroid lipofuscinosis with onset at birth or early childhood. Neuronal ceroid lipofuscinoses are progressive neurodegenerative, lysosomal storage diseases characterized by intracellular accumulation of autofluorescent liposomal material, and clinically by seizures, dementia, visual loss, and/or cerebral atrophy.Note: The disease is caused by mutations affecting the gene represented in this entry.
