Principle of the Assay: This assay employs a two-site sandwich ELISA to quantitate CASP8AP2 in samples. An antibody specific for CASP8AP2 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyCASP8AP2 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for CASP8AP2 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of CASP8AP2 bound in the initial step. The color development is stopped and the intensity of the color is measured.

CASP8AP2: Participates in TNF-alpha-induced blockade of glucocorticoid receptor (GR) transactivation at the nuclear receptor coactivator level, upstream and independently of NF- kappa-B. Suppresses both NCOA2- and NCOA3-induced enhancement of GR transactivation. Involved in TNF-alpha-induced activation of NF-kappa-B via a TRAF2-dependent pathway. Acts as a downstream mediator for CASP8-induced activation of NF-kappa-B. Required for the activation of CASP8 in FAS-mediated apoptosis. Required for histone gene transcription and progression through S phase.
