Principle of the Assay: This assay employs a two-site sandwich ELISA to quantitate CASP6 in samples. An antibody specific for CASP6 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyCASP6 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for CASP6 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of CASP6 bound in the initial step. The color development is stopped and the intensity of the color is measured.

NCBI Description

This gene encodes a member of the cysteine proteases that plays important roles in regulating apoptosis and neurodegeneration. The encoded protein is involved in the transmission of pain and axonal degeneration. Genetic deletion of this gene in mice results in the delay of axon pruning and protects from axon degeneration. CASP6: Involved in the activation cascade of caspases responsible for apoptosis execution. Cleaves poly(ADP-ribose) polymerase in vitro, as well as lamins. Overexpression promotes programmed cell death. Heterotetramer that consists of two anti-parallel arranged heterodimers, each one formed by a 18 kDa (p18) and a 11 kDa (p11) subunit. Interacts with BIRC6/bruce. Activation is suppressed by phosphorylation at Ser-257. Belongs to the peptidase C14A family. 2 isoforms of the human protein are produced by alternative splicing.
