Specificity
This assay has high sensitivity and excellent specificity for detection of CASP11. No significant cross-reactivity or interference between CASP11 and analogues was observed. NOTE: Limited by current skills and knowledge, it is impossible for us to complete the cross-reactivity detection between CASP11 and all the analogues, therefore, cross reaction may still exist in some cases.

Intended Uses: This CASP11 ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Human CASP11. This ELISA kit for research use only, not for therapeutic or test applications!

Principle of the Assay: CASP11 ELISA kit applies the quantitative sandwich enzyme immunoassay technique. The microtiter plate has been pre-coated with a monoclonal antibody specific for CASP11. Standards or samples are then added to the microtiter plate wells and CASP11 if present, will bind to the antibody pre-coated wells. In order to quantitatively determine the amount of CASP11 present in the sample, a standardized preparation of horseradish peroxidase (HRP)-conjugated polyclonal antibody, specific for CASP11 are added to each well to "sandwich" the CASP11 immobilized on the plate. The microtiter plate undergoes incubation, and then the wells are thoroughly washed to remove all unbound components. Next, substrate solutions are added to each well. The enzyme (HRP) and substrate are allowed to react over a short incubation period. Only those wells that contain CASP11 and enzyme-conjugated antibody will exhibit a change in color. The enzyme-substrate reaction is terminated by addition of a sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450 nm. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The CASP11 concentration in each sample is interpolated from this standard curve.

NCBI Description

The protein encoded by this gene belongs to the membrane-associated guanylate kinase (MAGUK) family, a class of proteins that functions as molecular scaffolds for the assembly of multiprotein complexes at specialized regions of the plasma membrane. This protein is also a member of the CARD protein family, which is defined by carrying a characteristic caspase-associated recruitment domain (CARD). This protein has a domain structure similar to that of CARD14 protein. The CARD domains of both proteins have been shown to specifically interact with BCL10, a protein known to function as a positive regulator of cell apoptosis and NF-kappaB activation. When expressed in cells, this protein activated NF-kappaB and induced the phosphorylation of BCL10. [provided by RefSeq, Jul 2008]

Uniprot Description

CARD11: a CARD (caspase recruitment domain) protein that belongs to the membrane-associated guanylate kinase (MAGUK) family. MAGUK proteins function as molecular scaffolds for the assembly of multiprotein complexes at specialized regions of the plasma membrane. Contains an N-terminal CARD domain, a central coiled-coil domain, and a C-terminal region containing a PDZ, an SH3, and a GUK. Its CARD domain associates with the CARD domain of BCL10. BCL10 activates NF-kappaB through the IkappaB kinase complex in response to upstream stimuli. Apparently activates NF-kappaB and induce the phosphorylation of BCL10. Detected in adult peripheral blood leukocytes, thymus, spleen and liver. Also found in promyelocytic leukemia HL-60 cells, chronic myelogenous leukemia K562 cells, Burkitt's lymphoma Raji cells and colorectal adenocarcinoma SW480 cells.

Product Notes

The Human CASP11 card11 (Catalog #AAA729646) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA729646 ELISA Kit recognizes Human CASP11. It is sometimes possible for the material contained within the vial of "Caspase 11, ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
