Intended Uses: This MDA ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Rat MDA. This ELISA kit for research use only!

Principle of the Assay: MDA ELISA kit applies the competitive enzyme immunoassay technique utilizing a polyclonal anti-MDA antibody and an MDA-HRP conjugate. The assay sample and buffer are incubated together with MDA-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the MDA concentration since MDA from samples and MDA-HRP conjugate compete for the anti-MDA antibody binding site. Since the number of sites is limited, as more sites are occupied by MDA from the sample, fewer sites are left to bind MDA-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The MDA concentration in each sample is interpolated from this standard curve.

NCBI Description

This gene encodes a cytosolic, NADP-dependent enzyme that generates NADPH for fatty acid biosynthesis. The activity of this enzyme, the reversible oxidative decarboxylation of malate, links the glycolytic and citric acid cycles. The regulation of expression for this gene is complex. Increased expression can result from elevated levels of thyroid hormones or by higher proportions of carbohydrates in the diet. [provided by RefSeq, Jul 2008]

ME1: a ubiquitous lipogenic enzyme of the malic enzymes family. Converts (S)-malate + NADP+ ?? pyruvate + CO2 + NADPH. Levels of ME1 and ATP-citrate lyase mRNA, another lipogenic enzyme, are coordinately increased 5-7 fold during the differentiation of 3T3-L1 preadipocytes into adipocytes. Up-regulated by short-term dietary restriction in mice. The cycling of pyruvate by ME1 generates cytosolic NADPH, whereas mitochondrial ME2 responds to elevated amino acids and serves to supply sufficient pyruvate for increased Krebs cycle flux when glucose is limiting.
