Principle of the Assay||FCGR3A ELISA kit applies the competitive enzyme immunoassay technique utilizing a polyclonal anti-FCGR3A antibody and an FCGR3A-HRP conjugate. The assay sample and buffer are incubated together with FCGR3A-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the FCGR3A concentration since FCGR3A from samples and FCGR3A-HRP conjugate compete for the anti-FCGR3A antibody binding site. Since the number of sites is limited, as more sites are occupied by FCGR3A from the sample, fewer sites are left to bind FCGR3A-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The FCGR3A concentration in each sample is interpolated from this standard curve.

This gene encodes a receptor for the Fc portion of immunoglobulin G, and it is involved in the removal of antigen-antibody complexes from the circulation, as well as other other antibody-dependent responses. This gene (FCGR3A) is highly similar to another nearby gene (FCGR3B) located on chromosome 1. The receptor encoded by this gene is expressed on natural killer (NK) cells as an integral membrane glycoprotein anchored through a transmembrane peptide, whereas FCGR3B is expressed on polymorphonuclear neutrophils (PMN) where the receptor is anchored through a phosphatidylinositol (PI) linkage. Mutations in this gene have been linked to susceptibility to recurrent viral infections, susceptibility to systemic lupus erythematosus, and alloimmune neonatal neutropenia.
