Intended Uses: This CRKL ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Human CRKL. This ELISA kit for research use only!

Principle of the Assay: CRKL ELISA kit applies the competitive enzyme immunoassay technique utilizing a polyclonal anti-CRKL antibody and an CRKL-HRP conjugate. The assay sample and buffer are incubated together with CRKL-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the CRKL concentration since CRKL from samples and CRKL-HRP conjugate compete for the anti-CRKL antibody binding site. Since the number of sites is limited, as more sites are occupied by CRKL from the sample, fewer sites are left to bind CRKL-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The CRKL concentration in each sample is interpolated from this standard curve.

NCBI Description

This gene encodes a member of the type I cytokine receptor family. The encoded protein is a receptor for thymic stromal lymphopoietin (TSLP). Together with the interleukin 7 receptor (IL7R), the encoded protein and TSLP activate STAT3, STAT5, and JAK2 pathways, which control processes such as cell proliferation and development of the hematopoietic system. Rearrangement of this gene with immunoglobulin heavy chain gene (IGH) on chromosome 14, or with P2Y purinoceptor 8 gene (P2RY8) on the same X or Y chromosomes is associated with B-progenitor acute lymphoblastic leukemia (ALL) and Down syndrome ALL. Alternatively spliced transcript variants have been found for this gene. [provided by RefSeq, Sep 2014]
