This assay has high sensitivity and excellent specificity for detection of CYP2D6. No significant cross-reactivity or interference between CYP2D6 and analogues was observed. NOTE: Limited by current skills and knowledge, it is impossible for us to complete the cross-reactivity detection between CYP2D6 and all the analogues, therefore, cross reaction may still exist in some cases.

Intended Uses: This CYP2D6 ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Canine CYP2D6. This ELISA kit for research use only, not for therapeutic or test applications!

Principle of the Assay: CYP2D6 ELISA kit applies the competitive enzyme immunoassay technique utilizing an anti-CYP2D6 antibody and an CYP2D6-HRP conjugate. The assay sample and buffer are incubated together with CYP2D6-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the CYP2D6 concentration since CYP2D6 from samples and CYP2D6-HRP conjugate compete for the anti-CYP2D6 antibody binding site. Since the number of sites is limited, as more sites are occupied by CYP2D6 from the sample, fewer sites are left to bind CYP2D6-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The CYP2D6 concentration in each sample is interpolated from this standard curve.

NCBI Description

This gene encodes a member of the cytochrome P450 superfamily of enzymes. The cytochrome P450 proteins are monooxygenases which catalyze many reactions involved in drug metabolism and synthesis of cholesterol, steroids and other lipids. This protein localizes to the endoplasmic reticulum and is known to metabolize as many as 25% of commonly prescribed drugs. Its substrates include antidepressants, antipsychotics, analgesics and antitussives, beta adrenergic blocking agents, antiarrythmics and antiemetics. The gene is highly polymorphic in the human population; certain alleles result in the poor metabolizer phenotype, characterized by a decreased ability to metabolize the enzyme's substrates. Some individuals with the poor metabolizer phenotype have no functional protein since they carry 2 null alleles whereas in other individuals the gene is absent. This gene can vary in copy number and individuals with the ultrarapid metabolizer phenotype can have 3 or more active copies of the gene. Alternatively spliced transcript variants encoding different isoforms have been found for this gene. [provided by RefSeq, Jul 2014]
