Principle of the Assay: CD1D ELISA kit applies the competitive enzyme immunoassay technique utilizing an anti-CD1D antibody and an CD1D-HRP conjugate. The assay sample and buffer are incubated together with CD1D-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the CD1D concentration since CD1D from samples and CD1D-HRP conjugate compete for the anti-CD1D antibody binding site. Since the number of sites is limited, as more sites are occupied by CD1D from the sample, fewer sites are left to bind CD1D-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The CD1D concentration in each sample is interpolated from this standard curve.

This gene encodes a divergent member of the CD1 family of transmembrane glycoproteins, which are structurally related to the major histocompatibility complex (MHC) proteins and form heterodimers with beta-2-microglobulin. The CD1 proteins mediate the presentation of primarily lipid and glycolipid antigens of self or microbial origin to T cells. The human genome contains five CD1 family genes organized in a cluster on chromosome 1. The CD1 family members are thought to differ in their cellular localization and specificity for particular lipid ligands. The protein encoded by this gene localizes to late endosomes and lysosomes via a tyrosine-based motif in the cytoplasmic tail.
