Intended Uses: This C1QAb ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Mouse C1QAb. This ELISA kit for research use only, not for therapeutic or test applications!

Principle of the Assay: C1QAb ELISA kit applies the competitive enzyme immunoassay technique utilizing Complement 1q antigen and an C1QAb-HRP conjugate. The assay sample and buffer are incubated together with C1QAb-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the C1QAb concentration since C1QAb from samples and C1QAb-HRP conjugate compete for the Complement 1q antigen binding site. Since the number of sites is limited, as more sites are occupied by C1QAb from the sample, fewer sites are left to bind C1QAb-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The C1QAb concentration in each sample is interpolated from this standard curve.

This gene encodes a member of a family of proteins that function as components of basement membranes and may play a role in cell adhesion. Mutations in this gene have been associated with late-onset retinal degeneration. The protein may be encoded by either a bicistronic transcript including sequence from the upstream membrane frizzled-related protein gene (MFRP), or by a monocistronic transcript expressed from an internal promoter. LORD is an autosomal dominant disorder characterized by onset in the fifth to sixth decade with night blindness and punctate yellow-white deposits in the retinal fundus, progressing to severe central and peripheral degeneration, with choroidal neovascularization and chorioretinal atrophy.
