Principle of the assay: C/EBP? ELISA kit applies the quantitative sandwich enzyme immunoassay technique. The microtiter plate has been pre-coated with a monoclonal antibody specific for C/EBP?. Standards or samples are then added to the microtiter plate wells and C/EBP? if present, will bind to the antibody pre-coated wells. In order to quantitatively determine the amount of C/EBP? present in the sample, a standardized preparation of horseradish peroxidase (HRP)-conjugated polyclonal antibody, specific for C/EBP? are added to each well to “sandwich” the C/EBP? immobilized on the plate. The microtiter plate undergoes incubation, and then the wells are thoroughly washed to remove all unbound components. Next, substrate solutions are added to each well. The enzyme (HRP) and substrate are allowed to react over a short incubation period. Only those wells that contain C/EBP? and enzyme-conjugated antibody will exhibit a change in color. The enzyme-substrate reaction is terminated by addition of a sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450 nm. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The C/EBP? concentration in each sample is interpolated from this standard curve.

NCBI Description

This intronless gene encodes a transcription factor that contains a basic leucine zipper (bZIP) domain. The encoded protein functions as a homodimer but can also form heterodimers with CCAAT/enhancer-binding proteins alpha, delta, and gamma. Activity of this protein is important in the regulation of genes involved in immune and inflammatory responses, among other processes. The use of alternative in-frame AUG start codons results in multiple protein isoforms, each with distinct biological functions. [provided by RefSeq, Oct 2013]

C/EBP-beta: a bZIP transcription factor which can form homodimers or heterodimers with the related proteins CEBP-alpha, -delta and -gamma. Involved in immune and inflammatory responses. Specifically binds to regulatory regions of genes encoding IL-6, other cytokines and several acute-phase proteins. There are two forms of C/EBPbeta, the 38kDa liver activating protein (LAP) and the 20kDa liver inhibitory protein (LIP) which may be products of alternative translation. LAP is a transcriptional activator while LIP may inhibit C/EBPbeta transcriptional activity. Phosphorylated and activated by ERK1/2.
