Bovine RE1-Silencing Transcription Factor (REST) ELISA Kit

Principle of the Assay: REST ELISA kit applies the competitive enzyme immunoassay technique utilizing an anti-REST antibody and an REST-HRP conjugate. The assay sample and buffer are incubated together with REST-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the REST concentration since REST from samples and REST-HRP conjugate compete for the anti-REST antibody binding site. Since the number of sites is limited, as more sites are occupied by REST from the sample, fewer sites are left to bind REST-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The REST concentration in each sample is interpolated from this standard curve.

This gene was initially identified as a transcriptional repressor that represses neuronal genes in non-neuronal tissues. However, depending on the cellular context, this gene can act as either an oncogene or a tumor suppressor. The encoded protein is a member of the Kruppel-type zinc finger transcription factor family. It represses transcription by binding a DNA sequence element called the neuron-restrictive silencer element. The protein is also found in undifferentiated neuronal progenitor cells and it is thought that this repressor may act as a master negative regulator of neurogenesis. Transcriptional repression by REST-CDYL via the recruitment of histone methyltransferase EHMT2 may be important in transformation suppression. Key repressor of gene expression in hypoxia; represses genes in hypoxia by direct binding to an RE1/NRSE site on their promoter regions (PubMed:27531581).
