Intended Uses: This PSMD2 ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Bovine PSMD2. This ELISA kit for research use only!

Principle of the Assay: PSMD2 ELISA kit applies the competitive enzyme immunoassay technique utilizing a Polyclonal anti-PSMD2 antibody and an PSMD2-HRP conjugate. The assay sample and buffer are incubated together with PSMD2-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the PSMD2 concentration since PSMD2 from samples and PSMD2-HRP conjugate compete for the anti-PSMD2 antibody binding site. Since the number of sites is limited, as more sites are occupied by PSMD2 from the sample, fewer sites are left to bind PSMD2-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The PSMD2 concentration in each sample is interpolated from this standard curve.

NCBI Description

The 26S proteasome is a multicatalytic proteinase complex with a highly ordered structure composed of 2 complexes, a 20S core and a 19S regulator. The 20S core is composed of 4 rings of 28 non-identical subunits; 2 rings are composed of 7 alpha subunits and 2 rings are composed of 7 beta subunits. The 19S regulator is composed of a base, which contains 6 ATPase subunits and 2 non-ATPase subunits, and a lid, which contains up to 10 non-ATPase subunits. Proteasomes are distributed throughout eukaryotic cells at a high concentration and cleave peptides in an ATP/ubiquitin-dependent process in a non-lysosomal pathway. An essential function of a modified proteasome, the immunoproteasome, is the processing of class I MHC peptides. This gene encodes one of the non-ATPase subunits of the 19S regulator lid. In addition to participation in proteasome function, this subunit may also participate in the TNF signalling pathway since it interacts with the tumor necrosis factor type 1 receptor. A pseudogene has been identified on chromosome 1. Alternative splicing results in multiple transcript variants of this gene. [provided by RefSeq, Jul 2013]
