Testing Data (Image: Western blot of crude extract of the human carcinoma HeLa cell line. Lane 15 was probed with Actin, our monoclonal antibody to all six actin isotypes, giving a strong clean band at an SDS-PAGE molecular weight of 42kDa. lane 16 was probed with our antibody to 14-3-3eta (14-3-3 eta), which has an SDS-PAGE molecular weight of 28kDa. Lanes 17 was probed with our monoclonal antibody to vimentin. Protocol on Datasheet.)

(Image: HeLa cells stained with Actin (red) and also with our chicken polyclonal antibody to Vimentin, (green). The actin antibody stains the submembranous actin rich cytoskeleton and also stress fibers, bundles of actin associated with adhesion sites. The vimentin antibody stains a quite different cytoskeletal network, the intermediate filaments. The blue stain reveals DNA in the nuclei of these cells. Protocol on Datasheet.)

Related Product Information for anti-Actin antibody
Actin is one of the most abundant and highly conserved proteins of eukaryotes. Mammalian actins are the product of six different genes with differing distribution patterns in cell types and in tissues. The molecular weight of all six proteins is 42kDa, and one or more actins is found in essentially every type of crude cellular and tissue extract. Actin is regarded as a "house keeping" protein which is generally not altered much in expression as a result of experimental manipulations. So quantitation of the actin band on the western blots is used as a standard against which the band density of other proteins can be compared. Glyceraldehyde 3 phosphate dehydrogenase (GAPDH) is another "house keeping" protein used in this way. Since our antibody binds all six isotypes very strongly on western blots, it is a very effective blotting standard. It also works in immunocytochemical experiments, binding stongly and cleanly to filopodia, membrane ruffles and stress fibers, all known to be rich in actin

Product Notes

The Actin (Catalog #AAA555035) is an Antibody and is intended for research purposes only. The product is available for immediate purchase. The Actin, Monoclonal Antibody, Mouse reacts with Bovine, Human, Mouse, Porcine, Rat and may cross-react with other species as described in the data sheet. AAA Biotech's Actin can be used in a range of immunoassay formats including, but not limited to, Immunofluorescence (IF), Western Blot (WB). Researchers should empirically determine the suitability of the Actin for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "Actin, Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
