MBD3 blocking peptide

MBD3 Antibody (N-term) Blocking peptide

Synonyms
MBD3; MBD3 Antibody (N-term) Blocking peptide; Methyl-CpG-binding domain protein 3; Methyl-CpG-binding protein MBD3; MBD3 blocking peptide
Ordering
Specificity
The synthetic peptide sequence used to generate the antibody was selected from the N-term region of MBD3. A 10 to 100 fold molar excess to antibody is recommended. Precise conditions should be optimized for a particular assay.
Form/Format
Synthetic peptide was lyophilized with 100% acetonitrile and is supplied as a powder. Reconstitute with 0.1 ml DI water for a final concentration of 1 mg/ml.
Sequence Length
291
Cellular Location
Nucleus. Chromosome. Note: Nuclear, in discrete foci. Detected on chromatin, at promoter regions of active genes
Preparation and Storage
Maintain refrigerated at 2-8 degree C for up to 6 months. For long term storage store at -20 degree C.
Related Product Information for MBD3 blocking peptide
Acts as transcriptional repressor and plays a role in gene silencing. Does not bind to DNA by itself (PubMed:12124384). Binds to DNA with a preference for sites containing methylated CpG dinucleotides (in vitro). Binds to a lesser degree DNA containing unmethylated CpG dinucleotides (PubMed:24307175). Recruits histone deacetylases and DNA methyltransferases.

NCBI Description

DNA methylation is the major modification of eukaryotic genomes and plays an essential role in mammalian development. This gene belongs to a family of nuclear proteins which are characterized by the presence of a methyl-CpG binding domain (MBD). The encoded protein is a subunit of the NuRD, a multisubunit complex containing nucleosome remodeling and histone deacetylase activities. Unlike the other family members, the encoded protein is not capable of binding to methylated DNA. The protein mediates the association of metastasis-associated protein 2 with the core histone deacetylase complex. Alternative splicing results in multiple transcript variants of this gene. [provided by RefSeq, Jul 2013]

Uniprot Description

MBD3: methyl-CpG binding (MBD) proteins appear to bind differentially methylated domains (DMDs) of DNA, to recruit repression complexes, and to silence the locus. The methyl CpG binding domain of human MBD3 may not directly bind mCpG but does interact with the NuRD/Mi2 components HDAC1 and MTA2. Plays a role in histone deacetylation and nucleosome remodeling. Increased levels have been reported in lung cancer and glioblastomas and may be a therapeutic target.

Product Notes

The MBD3 mbd3 (Catalog #AAA9220166) is a Blocking Peptide and is intended for research purposes only. The product is available for immediate purchase. It is sometimes possible for the material contained within the vial of "MBD3, Blocking Peptide" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
