BRDT blocking peptide

BRDT Antibody (N-term) Blocking Peptide

Gene Names
BRDT; CT9; BRD6
Synonyms
BRDT; BRDT Antibody (N-term) Blocking Peptide; Bromodomain testis-specific protein; Cancer/testis antigen 9; CT9; RING3-like protein; BRDT blocking peptide
Ordering
Specificity
The synthetic peptide sequence used to generate the antibody was selected from the N-term region of human BRDT. A 10 to 100 fold molar excess to antibody is recommended. Precise conditions should be optimized for a particular assay.
Form/Format
The synthetic peptide was lyophilized with 100% acetonitrile and is supplied as a powder. Reconstitute with 0.1 ml deionized water for a final concentration of 1 mg/ml.
Sequence Length
947
Cellular Location
Nucleus Note: Detected on chromatin
Tissue Location
Testis-specific. A 3-fold higher expression is seen in adult testis than in embryo testis. Expression seems to be correlated with histone H4 hyperacetylation during the haploid phase of spermatogenesis (spermiogenesis). No expression, or very low expression is seen in patients' testes with abnormal spermatogenesis. Expressed in cancers such as non-small cell lung cancer and squamous cell carcinomas of the head and neck as well as of esophagus, but not in melanoma or in cancers of the colon, breast, kidney and bladder.
Preparation and Storage
Maintain refrigerated at 2-8 degree C for up to 6 months. For long term storage store at -20 degree C.
Related Product Information for BRDT blocking peptide
Testis-specific chromatin protein that specifically binds histone H4 acetylated at 'Lys-5' and 'Lys-8' (H4K5ac and H4K8ac, respectively) and plays a key role in spermatogenesis. Required in late pachytene spermatocytes: plays a role in meiotic and post-meiotic cells by binding to acetylated histones at the promoter of specific meiotic and post-meiotic genes, facilitating their activation at the appropriate time. In the post-meiotic phase of spermatogenesis, binds to hyperacetylated histones and participates in their general removal from DNA. Also acts as a component of the splicing machinery in pachytene spermatocytes and round spermatids and participates in 3'-UTR truncation of specific mRNAs in post-meiotic spermatids. Required for chromocenter organization, a structure comprised of peri-centromeric heterochromatin.

NCBI Description

BRDT is similar to the RING3 protein family. It possesses 2 bromodomain motifs and a PEST sequence (a cluster of proline, glutamic acid, serine, and threonine residues), characteristic of proteins that undergo rapid intracellular degradation. The bromodomain is found in proteins that regulate transcription. Several transcript variants encoding multiple isoforms have been found for this gene. [provided by RefSeq, Jun 2011]

Uniprot Description

BRDT: an atypical protein kinase of the BRD family. Possesses 2 bromodomain motifs and a PEST sequence (a cluster of proline, glutamic acid, serine, and threonine residues), characteristic of proteins that undergo rapid intracellular degradation. The bromodomain is found in proteins that regulate transcription. A testis-specific protein that is also expressed at a high level in non-small cell lung cancer

Protein type: Cancer Testis Antigen (CTA); Protein kinase, Ser/Thr (non-receptor); Protein kinase, atypical; Kinase, protein; ATYPICAL group; BRD family

Chromosomal Location of Human Ortholog: 1p22.1

Cellular Component: nucleus

Molecular Function: histone binding; transcription coactivator activity

Biological Process: chromatin remodeling; male meiosis; male meiosis I; positive regulation of transcription, meiotic; regulation of RNA splicing; spermatogenesis

Product Notes

The BRDT brdt (Catalog #AAA9226601) is a Blocking Peptide and is intended for research purposes only. The product is available for immediate purchase. It is sometimes possible for the material contained within the vial of "BRDT, Blocking Peptide" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
