Principle of the Assay: The microtiter plate provided in this kit has been pre-coated with an antibody specific to Bcl-2-binding component 3. Standards or samples are then added to the appropriate microtiter plate wells with a biotin-conjugated polyclonal antibody preparation specific for Bcl-2-binding component 3 and Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. Then a TMB substrate solution is added to each well. Only those wells that contain Bcl-2-binding component 3, biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of a sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450 nm +/- 2 nm. The concentration of Bcl-2-binding component 3 in the samples is then determined by comparing the O.D. of the samples to the standard curve.

Uniprot Description

PUMA: Essential mediator of p53/TP53-dependent and p53/TP53- independent apoptosis. Interacts with MCL1 and BCL2A1. Interacts with BCL2 and BCL2L1/BCL-XL. By DNA damage, glucocorticoid treatment, growth factor deprivation and p53/TP53. Ubiquitously expressed. Belongs to the Bcl-2 family. 4 isoforms of the human protein are produced by alternative splicing.

Protein type: Mitochondrial

Cellular Component: endoplasmic reticulum; lysosome; mitochondrion

Molecular Function: ATPase binding; glycoprotein binding; protein binding

Biological Process: apoptosis; brain development; caspase activation; determination of adult life span; DNA damage response, signal transduction by p53 class mediator resulting in induction of apoptosis; glial cell differentiation; negative regulation of growth; positive regulation of apoptosis; positive regulation of neuron apoptosis; positive regulation of protein homooligomerization; reduction of endoplasmic reticulum calcium ion concentration; release of cytochrome c from mitochondria; release of sequestered calcium ion into cytosol; response to DNA damage stimulus

Product Notes

The Rat Bbc3 bbc3 (Catalog #AAA288278) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA288278 ELISA Kit recognizes Rat Bbc3. It is sometimes possible for the material contained within the vial of "Bcl-2-binding component 3, ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
