Benefits
The Quick ELISA kits, assay takes less than 1.5 hours. Detect Human Azurocidin with <10pg/ml sensitivity. Format: 96-well plate with removable strips.
This is a TMB colorimetric sandwich ELISA kit with short assay time and quick experiment set up.
Principle
Human AZU1 Pre-Coated ELISA (Enzyme-Linked Immunosorbent Assay) kit is a solid phase immunoassay specially designed to measure Human AZU1 in cell culture supernatants, cell lysates, serum and plasma (heparin, EDTA). It uses our proprietary Quick ELISA technology. Quick ELISA technology employs capture antibodies conjugated to an affinity tag that is recognized by the polyclonal antibody used to coat our Quick ELISA plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. The kit contains recombinant Human AZU1 with immunogen: Expression system for standard: NS0; Immunogen sequence: I27-P248.

To measure Human AZU1, add standards and samples to the wells, then add antibody cocktail. Wash the wells with PBS or TBS buffer, and add TMB. TMB is substrate to HRP and will be catalyzed to produce a blue color product, which changes into yellow after adding acidic stop solution. The density of the yellow product is linearly proportional to Human AZU1 in the sample. Read the density of the yellow product in each well using a plate reader, and benchmark the sample wells' readings against the standard curve to determine the concentration of Human AZU1 in the sample.

Preparation and Storage
Store at 4 degree C for 6 months, at -20 degree C for 12 months. Avoid multiple freeze-thaw cycles. (Shipped with wet ice.)

Related Product Information for AZU1 elisa kit
Azurocidin, also known as cationic antimicrobial protein CAP37 or heparin-binding protein (HBP), is a protein that in humans is encoded by the AZU1 gene. This encoded protein is a member of the serine protease gene family, but it is not a serine proteinase, because the active site serine and histidine residues are replaced. Azurocidin is mapped to 19p13. 3. The protein encoded by this gene is an azurophil granule antibiotic protein, with antibacterial activity. It is also an important multifunctional inflammatory mediator. In addition to it, Azurocidin is also a specific chemoattractant for monocytes. It lacks the chemotactic activity for neutrophils and lymphocytes, and this gene is probably responsible for the wave of monocytes that follows the initial wave of PMNs typical of the inflammatory response.

NCBI Description

Azurophil granules, specialized lysosomes of the neutrophil, contain at least 10 proteins implicated in the killing of microorganisms. The protein encoded by this gene is an azurophil granule antibiotic protein, with monocyte chemotactic and antibacterial activity. It is also an important multifunctional inflammatory mediator. This encoded protein is a member of the serine protease gene family but it is not a serine proteinase, because the active site serine and histidine residues are replaced. The genes encoding this protein, neutrophil elastase 2, and proteinase 3 are in a cluster located at chromosome 19pter. All 3 genes are expressed coordinately and their protein products are packaged together into azurophil granules during neutrophil differentiation. [provided by RefSeq, Jul 2008]

Uniprot Description

AZU1: This is a neutrophil granule-derived antibacterial and monocyte- and fibroblast-specific chemotactic glycoprotein. Binds heparin. The cytotoxic action is limited to many species of Gram- negative bacteria; this specificity may be explained by a strong affinity of the very basic N-terminal half for the negatively charged lipopolysaccharides that are unique to the Gram-negative bacterial outer envelope. It may play a role in mediating recruitment of monocytes in the second wave of inflammation. Has antibacterial activity against the Gram-nagative bacterium P.aeruginosa, this activity is inhibited by LPS from P.aeruginosa. Acting alone, it does not have antimicrobial activity against the Gram-negative bacteria A.actinomycetemcomitans ATCC 29532, A.actinomycetemcomitans NCTC 9709, A.actinomycetemcomitans FDC-Y4, H.aphrophilus ATCC 13252, E.corrodens ATCC 23834, C.sputigena ATCC 33123, Capnocytophaga sp ATCC 33124, Capnocytophaga sp ATCC 27872 or E.coli ML-35. Has antibacterial activity against C.sputigena ATCC 33123 when acting synergistically with either elastase or cathepsin G. Belongs to the peptidase S1 family. Elastase subfamily.

Product Notes

The Human AZU1 azu1 (Catalog #AAA1753948) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA1753948 ELISA Kit recognizes Human AZU1. It is sometimes possible for the material contained within the vial of "Azurocidin, ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
