Intended Uses: This ATF6 ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Human ATF6. This ELISA kit for research use only, not for therapeutic or test applications!

Principle of the Assay: ATF6 ELISA kit applies the competitive enzyme immunoassay technique utilizing an anti-ATF6 antibody and an ATF6-HRP conjugate. The assay sample and buffer are incubated together with ATF6-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the ATF6 concentration since ATF6 from samples and ATF6-HRP conjugate compete for the anti-ATF6 antibody binding site. Since the number of sites is limited, as more sites are occupied by ATF6 from the sample, fewer sites are left to bind ATF6-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The ATF6 concentration in each sample is interpolated from this standard curve.

NCBI Description

This gene encodes a transcription factor that activates target genes for the unfolded protein response (UPR) during endoplasmic reticulum (ER) stress. Although it is a transcription factor, this protein is unusual in that it is synthesized as a transmembrane protein that is embedded in the ER. It functions as an ER stress sensor/transducer, and following ER stress-induced proteolysis, it functions as a nuclear transcription factor via a cis-acting ER stress response element (ERSE) that is present in the promoters of genes encoding ER chaperones. This protein has been identified as a survival factor for quiescent but not proliferative squamous carcinoma cells. There have been conflicting reports about the association of polymorphisms in this gene with diabetes in different populations, but another polymorphism has been associated with increased plasma cholesterol levels. This gene is also thought to be a potential therapeutic target for cystic fibrosis. [provided by RefSeq, Aug 2011]
