Principle of the Assay: ATF1 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-ATF1 antibody and an ATF1-HRP conjugate. The assay sample and buffer are incubated together with ATF1-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the ATF1 concentration since ATF1 from samples and ATF1-HRP conjugate compete for the anti-ATF1 antibody binding site. Since the number of sites is limited, as more sites are occupied by ATF1 from the sample, fewer sites are left to bind ATF1-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The ATF1 concentration in each sample is interpolated from this standard curve.

NCBI Description

This gene encodes an activating transcription factor, which belongs to the ATF subfamily and bZIP (basic-region leucine zipper) family. It influences cellular physiologic processes by regulating the expression of downstream target genes, which are related to growth, survival, and other cellular activities. This protein is phosphorylated at serine 63 in its kinase-inducible domain by serine/threonine kinases, cAMP-dependent protein kinase A, calmodulin-dependent protein kinase I/II, mitogen- and stress-activated protein kinase and cyclin-dependent kinase 3 (cdk-3). Its phosphorylation enhances its transactivation and transcriptional activities, and enhances cell transformation. Fusion of this gene and FUS on chromosome 16 or EWSR1 on chromosome 22 induced by translocation generates chimeric proteins in angiomatoid fibrous histiocytoma and clear cell sarcoma. Plays an important role in the trans-activation of the MHC class II trans-activator (CIITA) promoter III in B cells.
